| 1 | Bavachin enhances NLRP3 inflammasome activation induced by ATP or nigericin and causes idiosyncratic hepatotoxicity显示文摘Psoraleae Fructus(PF)is a well-known traditional herbal medicine in China, and it is widely used for osteoporosis, vitiligo, and other diseases in clinical settings. However, liver injury caused by PF and its preparations has been frequently reported in recent years. Our previous studies have demonstrated that PF could cause idiosyncratic drug-induced liver injury(IDILI),but the mechanism underlying its hepatotoxicity remains unclear. This paper reports that bavachin isolated from PF enhances the specific stimuli-induced activation of the NLRP3 inflammasome and leads to hepatotoxicity.Bavachin boosts the secretion of IL-ip and caspase-1 caused by ATP or nigericin but not those induced by poly(I:C),monosodium urate crystal,or intracellular lipopolysaccharide.Bavachin does not affect AIM2 or NLRC4 inflammasome activation.Mechanistically, bavachin specifically increases the production of nigericin-induced mitochondrial reactive oxygen species among the most important upstream events in the activation of the NLRP3 inflammasome.Bavachin increases the levels of aspartate transaminase and alanine aminotransferase in serum and hepatocyte injury accompanied by the secretion of IL-ip via a mouse model of lipopolysaccharide-mediated susceptibility to IDILI.These results suggest that bavachin specifically enhances the ATP-or nigericin-induced activation of the NLRP3 inflammasome.Bavachin also potentially contributes to PF-induced idiosyncratic hepatotoxicity. Moreover, bavachin and PF should be evaded among patients with diseases linked to the ATP-or nigericin-mediated activation of the NLRP3 inflammasome, which may be a dangerous factor for liver injury. | Nan Qin Guang Xu Yan Wang Xiaoyan Zhan Yuan Gao Zhilei Wang Shubin Fu Wei Shi Xiaorong Hou Chunyu Wang Ruisheng Li Yan Liu Jiabo Wang Haiping Zhao Xiaohe Xiao Zhaofang Bai | 2021 | Frontiers of Medicine2021,15,4: | 9 |
| 2 | Numerical simulation of profile control by clay particles after polymer flooding显示文摘A three-dimensional,two-phase,five-component mathematical model has been developed to describe flow characteristics of clay particles and flocs in the profile control process,in which the clay particle suspension is injected into the formation to react with residual polymer. This model considers the reaction of clay particles with residual polymer, apparent viscosity of the mixture, retention of clay particles and flocs,as well as the decline in porosity and permeability caused by the retention of clay particles and flocs.A finite difference method is used to discretize the equation for each component in the model. The Runge-Kutta method is used to solve the polymer flow equation, and operator splitting algorithms are used to split the flow equation for clay particles into a hyperbolic equation for convection and a parabolic equation for diffusion, which effectively ensures excellent precision, high speed and good stability. The numerical simulation had been applied successfully in the 4-P1920 unit of the Lamadian Oilfield to forecast the blocking capacity of clay particle suspension and to optimize the injection parameters. | Feng Qihong Shi Shubin Wang Sen Zheng Lu | 2010 | Petroleum Science2010,7,4: | 2 |

| 7 | Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector. Some of JUNO's features make it an excellent location for^8B solar neutrino measurements, such as its low-energy threshold, high energy resolution compared with water Cherenkov detectors, and much larger target mass compared with previous liquid scintillator detectors. In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable, assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition, approximately 60,000 signal and 30,000 background events are expected. This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter, which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle. Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better, depending on the central value, and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND experiment. | 8 | GATA2^(−/−) human ESCs undergo attenuatedendothelial to hematopoietic transition andthereafter granulocyte commitment显示文摘Background: Hematopoiesis is a progressive process collectively controlled by an elaborate network of transcriptionfactors (TFs). Among these TFs, GATA2 has been implicated to be critical for regulating multiple steps of hematopoiesisin mouse models. However, whether similar function of GATA2 is conserved in human hematopoiesis, especially duringearly embryonic development stage, is largely unknown. Results: To examine the role of GATA2 in human background, we generated homozygous GATA2 knockout humanembryonic stem cells (GATA2^(−/−) hESCs) and analyzed their blood differentiation potential. Our results demonstratedthat GATA2^(−/−) hESCs displayed attenuated generation of CD34^(+)CD43^(+) hematopoietic progenitor cells (HPCs), due tothe impairment of endothelial to hematopoietic transition (EHT). Interestingly, GATA2^(−/−) hESCs retained the potentialto generate erythroblasts and macrophages, but never granulocytes. We further identified that SPI1 downregulationwas partially responsible for the defects of GATA2^(−/−) hESCs in generation of CD34^(+)CD43^(+) HPCs and granulocytes.Furthermore, we found that GATA2^(−/−) hESCs restored the granulocyte potential in the presence of Notch signaling. Conclusion: Our findings revealed the essential roles of GATA2 in EHT and granulocyte development throughregulating SPI1, and uncovered a role of Notch signaling in granulocyte generation during hematopoiesis modeled byhuman ESCs. | Ke Huang Juan Du Ning Ma Jiajun Liu Pengfei Wu Xiaoya Dong Minghui Meng Wenqian Wang Xin Chen Xi Shi Qianyu Chen Zhongzhou Yang Shubin Chen Jian Zhang Yuhang Li Wei Li Yi Zheng Jinglei Cai Peng Li Xiaofang Sun Jinyong Wang Duanqing Pei Guangjin Pan | 2015 | Cell Regeneration2015,4,1: | 0 |
| 9 | Harnessing the Unique Features of 2D Materials toward Dendrite-free Metal Anodes显示文摘Electrochemically active metal anodes, such as lithium, sodium, potassium, and zinc, have attracted great research interests in the advanced rechargeable batteries owing to their superior theoretical energy densities.Unfortunately, the metal anodes suffer from the huge volume changes with loss of active materials during the plating and stripping processes, resulting in fast capacity decay. Moreover, the random growth of dendrites on the metal anodes will penetrate the separator, causing severe safety issues.Engineering metal anodes by introducing the 2D materials are widely investigated to alleviate these issues.Benefitting from the ultrathin structure feature and unique electrical properties,2D materials are regarded as one of the best host of metal anodes.Besides, the tunable active sites on basal plane enable 2D materials to achieve favorable interaction with metal anodes. Moreover, some 2D materials exhibit good mechanical strength and flexibility, serving as building block for the artificial solid electrolyte interphase. In this review,we mainly disclosed the correlations between the intrinsic properties of 2D materials and their functions in guiding uniform nucleation, controlling the growth of metals, and accommodating the volume change. Also, the challenges of 2D materials in metal anodes are well discussed. Finally, the future directions to develop highperformance metal anodes by taking advantage of these unique features of 2D materials are proposed. | Zhenjiang Cao Yongzheng Zhang Yanglansen Cui Jianan Gu Zhiguo Du Yongzheng Shi Kai Shen Hao Chen Bin Li Shubin Yang | 2022 | Energy & Environmental Materials2022,5,1: | 0 |
