| 5 | Xinglou Chengqi Decoction improves neurological function in experimental stroke mice as evidenced by gut microbiota analysis and network pharmacology显示文摘The current study was designed to explore the brain protection mechanism of Xinglou Chengqi Decoction(XCD)based on gut microbiota analysis and network pharmacology. A transient middle cerebral artery occlusion(MCAO) model of mice was established, followed by behavioral evaluation, TTC and TUNEL staining. Additionally, to investigate the effects of gut microbiota on neurological function after stroke, C57BL/6 mice were treated with anti-biotic cocktails 14 days prior to ischemic stroke(IS) to deplete the gut microbiota. High-throughput 16S rDNA gene sequencing, metabonomics technique, and flow multifactor technology were used to analyze bacterial communities, SCFAs and inflammatory cytokines respectively. Finally, as a supplement, network pharmacology and molecular docking were applied to fully explore the multicomponent-multitarget-multichannel mechanism of XCD in treating IS, implicated in ADME screening, target identification, network analysis, functional annotation, and pathway enrichment analysis.We found that XCD effectively improved neurological function, relieved cerebral infarction and decreased the neuronal apoptosis.Moreover, XCD promoted the release of anti-inflammatory factor like IL-10, while down-regulating pro-inflammatory factors such as TNF-α, IL-17A, and IL-22. Furthermore, XCD significantly increased the levels of short chain fatty acids(SCFAs), especially butyric acid. The mechanism might be related to the regulation of SCFAs-producing bacteria like Verrucomicrobia and Akkermansia, and bacteria that regulate inflammation like Paraprevotella, Roseburia, Streptophyta and Enterococcu. Finally, in the network pharmacological analysis, 51 active compounds in XCD and 44 intersection targets of IS and XCD were selected. As a validation, components in XCD docked well with key targets. It was obviously that biological processes were mainly involved in the regulation of apoptotic process, inflammatory response, response to fatty acid, and regulation of establishment of endothelial barrier in GO enrichment. XCD can improve neurological function in experimental stroke mice, partly due to the regulation of gut microbiota. | 8 | Compromised small-world efficiency of structural brain networks in schizophrenic patients and their unaffected parents显示文摘Several lines of evidence suggest that efficient information integration between brain regions is disrupted in schizophrenia. Abnormalities in white matter tracts that interconnect brain regions may be directly relevant to this pathophysiological process. As a complex mental disorder with high heritability, mapping abnormalities in patients and their firstdegree relatives may help to disentangle the risk factors for schizophrenia. We established a weighted network model of white matter connections using diffusion tensor imaging in 25 nuclear families with schizophrenic probands(19 patients and 41 unaffected parents) and two unrelated groups of normal controls(24 controls matched with patients and 26 controls matched with relatives). The patient group showed lower global efficiency and local efficiency. The decreased regional efficiency was localized in hubs such as the bilateral frontal cortices, bilateral anterior cingulate cortices, and left precuneus. The global effi ciency was negatively correlated with cognition scores derived from a 5-factor model of schizophrenic psychopathology.We also found that unaffected parents displayed decreased regional efficiency in the right temporal cortices, left supplementary motor area, left superior temporal pole, and left thalamus. The global efficiency tended to be lower in unaffected parents. | 10 | Optimal Waist Circumference Cut-off values for Identifying Metabolic Risk Factors in Middle-aged and Elderly Subjects in Shandong Province of China显示文摘Objective To study the optimal waist circumference(WC) cut-off values for identifying metabolic risk factors in middle-aged and elderly subjects in Shandong Province of China. Methods A total of 2 873 men and 5 559 women were included in this cross-sectional study. Metabolic syndrome(MetS) was diagnosed according to the definition of Chinese Diabetes Society in 2004. The relation between WC and MetS was analyzed by multivariate logistic regression analysis. The optimal WC cut-off values were identified using the area under the ROC curve and the different diagnostic criteria for central obesity were compared. Results The WC was the risk factor for MetS independent of BMI, blood glucose, blood lipid, and blood pressure. The optimal WC cut-off value was 83.8 cm and 91.1 cm for identifying MetS in women and men, respectively. Compared with 80 cm and 85 cm for women and men, 85 cm and 90 cm had a higher Youden index for identifying all metabolic risk factors and MetS in women and men. | 11 | A good prognostic predictor for liver transplantation recipients with benign end-stage liver cirrhosis显示文摘BACKGROUND: Post-transplant model for predicting mortality(PMPM, calculated as-5.359+1.988×ln(serum creatinine [mg/d L])+1.089×ln(total bilirubin [mg/d L])) score has been proved to be a simple and accurate model for predicting the prognosis after liver transplantation(LT) in a single center study. Here we aim to verify this model in a large cohort of patients.METHODS: A total of 2727 patients undergoing LT with endstage liver cirrhosis from January 2003 to December 2010 were included in this retrospective study. Data were collected from the China Liver Transplant Registry(CLTR). PMPM score was calculated at 24-h and 7-d following LT. According to the PMPM score at 24-h, all patients were divided into the low-risk group(PMPM score ≤-1.4, n=2509) and the high-risk group(PMPM score >-1.4, n=218). The area under receiver operator characteristic curve(AUROC) was calculated for evaluating the prognostic accuracy.RESULTS: The 1-, 3-, and 5-year patient survival rates in the low-risk group were significantly higher than those in the high-risk group(90.23%, 88.01%, and 86.03% vs 63.16%, 59.62%, and 56.43%, respectively, P<0.001). In the high-risk group, 131 patients had a decreased PMPM score(≤-1.4) at 7-d, and their cumulative survival rate was significantly higher than the other 87 patients with sustained high PMPM score(>-1.4)(P<0.001). | 15 | Novel nanometer scaffolds regulate the biological behaviors of neural stem cells显示文摘Ideal tissue-engineered scaffold materials regulate proliferation, apoptosis and differentiation of cells seeded on them by regulating gene expression. In this study, aligned and randomly oriented collagen nanofiber scaffolds were prepared using electronic spinning technology. Their diameters and appearance reached the standards of tissue-engineered nanometer scaffolds. The nanofiber scaffolds were characterized by a high swelling ratio, high porosity and good mechanical properties. The proliferation of spinal cord-derived neural stem cells on novel nanofiber scaffolds was obviously enhanced. The proportions of cells in the S and G 2 /M phases noticeably increased. Moreover, the proliferation rate of neural stem cells on the aligned collagen nanofiber scaffolds was high. The expression levels of cyclin D1 and cyclin-dependent kinase 2 were increased. Bcl-2 expression was significantly increased, but Bax and caspase-3 gene expressions were obviously decreased. There was no significant difference in the differentiation of neural stem cells into neurons on aligned and randomly oriented collagen nanofiber scaffolds. These results indicate that novel nanofiber scaffolds could promote the proliferation of spinal cord-derived neural stem cells and inhibit apoptosis without inducing differentiation. | 17 | Berberine remodels adipose tissue to attenuate metabolic disorders by activating sirtuin 3显示文摘Adipose tissue remodelling is considered a critical pathophysiological hallmark of obesity and related metabolic diseases.Berberine(BBR),a natural isoquinoline alkaloid,has potent anti-hyperlipidaemic and anti-hyperglycaemic effects.This study aimed to explore the role of BBR in modulating adipose tissue remodelling and the underlying mechanisms.BBR protected high fat diet(HFD)-fed mice against adiposity,insulin resistance and hyperlipidemia.BBR alleviated adipose tissue inflammation and fibrosis by inhibiting macrophage infiltration,pro-inflammatory macrophage polarization and the abnormal deposition of extracellular matrix,and the effect was mediated by BBR directly binding and activating the deacetylase Sirtuin 3(SIRT3)and suppressing the activation of the mitogen-activated protein kinases and nuclear factor-κB signalling pathways.Furthermore,BBR decreased microRNA-155-5p secretion by macrophages,which in turn ameliorated liver injury.Moreover,BBR mitigated inflammatory responses in both LPS-stimulated macrophages and TNF-α-treated adipocytes and suppressed macrophage migration towards adipocytes by activating SIRT3.Collectively,this study revealed that BBR improved adipose tissue remodelling,and subsequently inhibited the secretion of microRNA-155-5p by macrophages,which alleviated adiposity,insulin resistance and liver injury in obese mice.The modulation of adipose tissue remodelling by activating SIRT3 could contribute to the anti-hyperlipidemic and anti-hyperglycemic effects of BBR. | Dan Li Chao Yang Jian-zhong Zhu Eduardo Lopez Tian Zhang Qiang Tong Cheng Peng Li-gen Lin | 2022 | Acta Pharmacologica Sinica2022,43,5: | 2 |
