| 8 | Junctional and somatic hypermutation-induced CX4C motif is critical for the recognition of a highly conserved epitope on HCV E2 by a human broadly neutralizing antibody显示文摘Induction of broadly neutralizing monoclonal antibodies(bNAbs)that bind to the viral envelope glycoproteins is a major goal of hepatitis C virus(HCV)vaccine research.The study of bNAbs arising in natural infection is essential in this endeavor.We generated a human antibody,8D6,recognizing the E2 protein of HCV isolated from a chronic hepatitis C patient.This antibody shows broadly neutralizing activity,which covers a pan-genotypic panel of cell culture-derived HCV virions(HCVcc).Functional and epitope analyses demonstrated that 8D6 can block the interaction between E2 and CD81 by targeting a highly conserved epitope on E2.We describe how the 8D6 lineage evolved via somatic hypermutation to achieve broad neutralization.We found that the V(D)J recombination-generated junctional and somatic hypermutation-induced disulfide bridge(C-C)motif in the CDRH3 is critical for the broad neutralization and binding activity of 8D6.This motif is conserved among a series of broadly neutralizing HCV antibodies,indicating a common binding model.Next,the 8D6 inferred germline(iGL)was reconstructed and tested for its binding affinity and neutralization activity.Interestingly,8D6 iGL-mediated relatively strong inhibition of the 1b genotype PR79L9 strain,suggesting that PR79L9 may serve as a potential natural viral strain that provides E2 sequences that induce bNAbs.Overall,our detailed epitope mapping and genetic studies of the HCV E2-specific mAb 8D6 have allowed for further refinement of antigenic sites on E2 and reveal a new mechanism to generate a functional CDRH3,while its iGL can serve as a probe to identify potential HCV vaccine strains. | Chunyan Yi Jing Xia Lan He Zhiyang Ling Xuesong Wang Yu Yan Jiangjun Wang Xinhao Zhao Weiguo Fan Xiaoyu Sun Ronghua Zhang Sheng Ye Rongguang Zhang Yongfen Xu Liyan Ma Yaguang Zhang Honglin Zhou Zhong Huang Junqi Niu Gang Long Junxia Lu Jin Zhong Bing Sun | 2021 | Cellular & Molecular Immunology2021,18,3: | 0 |
| 9 | Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND experiment. | 10 | A novel AH-D-A-type phase junction material to improve photovoltaic performance and device stability in fullerene OSCs显示文摘In order to boost power conversion efficiency(PCE) and operation stability of organic solar cells(OSCs),we propose a new idea of phase junction materials(PJMs) used as a photoactive layer component to improve device performance and stability.For this purpose,a novel PJM of H-TRC8 based on rhodanine unit was designed with a conjugated AH-D-A framework.Here,AH is a hydrogen-donating electron acceptor unit,D-A is an electron donor-acceptor unit.It is found that H-TRC8 has a good carriertransporting ability,as well as definite hydrogen-bond and D-A interaction with donor/acceptor materials.While H-TRC8 is added into the PBDB-T/PC60BM blend film with 1.0 vol% DIO(1,8-diiodooctane),the resulting blend film exhibited an enhanced absorption and improved morphology.The intermolecular hydrogen bond between H-TRC8 and PBDB-T plays an important role for them,which is confirmed via FT-IR spectra and 2D 1H NMR.As a result,the PBDB-T/PC60BM-based devices with 1.25 wt%H-TRC8 and 1.0 vol% DIO exhibit a significantly improved PCE of 8.06%,which is increased by 20.6% in comparison to that in the binary devices with 1.0 vol% DIO only(PCE=6.68%).Furthermore,the device stability is significantly enhanced with only 43% PCE roll-off at 150℃ for 120 h.This work indicates that AH-D-A-type PJMs are promising photovoltaic materials used as photoactive-layer components to achieve high-performance fullerene OSCs with high device stability. | Lin Shao Fei Tong Mengbing Zhu Hao Xia Wenhong Peng Hua Tan Bin Zhang Yu Liu Weiguo Zhu | 2020 | Chinese Chemical Letters2020,31,9: | 0 |
