2Intrinsic apoptotic pathway and G2/M cell cycle arrest involved in tubeimoside I-induced EC109 cell death显示文摘Objective: Squamous esophageal carcinoma is highly prevalent in developing countries, especially in China. Tu Bei Mu (TBM), a traditional folk medicine, has been used to treat esophageal squamous cell carcinoma (ESCC) for a long term. tubeimoside I (TBMS1) is the main component of TBM, exhibiting great anticancer potential. In this study, we investigated the mechanism of TBMS1 cytotoxic effect on EC109 cells. Methods: Comparative nuclear proteomic approach was applied in the current study and we identified several altered protein spots. Further biochemical studies were carried out to detect the mitochondrial membrane potential, cell cycle and corresponding proteins' expression and location. Results: Subcellular proteomic study in the nucleus from EC109 cells revealed that altered proteins were associated with mitochondrial function and cell proliferation. Further biochemical studies showed that TBMS1-induced molecular events were related to mitochondria-induced intrinsic apoptosis and P21-cyclin B1/cdc2 complex-related G2/M cell cycle arrest. 3Three-dimensional multimodal sub-diffraction imaging with spinning-disk confocal microscopy using blinking/ fluctuating probes显示文摘Three-dimensional imaging cannot be achieved easily using previously developed localization super-resolution techniques. Here, we present a three-dimensional multimodal sub-diffraction imaging technique with spinning-disk (SD) confocal microscopy called 3D-MUSIC, which not only has all the advantages of SD confocal microscopy such as fast imaging speed, high signal-to-noise ratio, and optical-sectioning capability, but also extends its spatial resolution limit along all three dimensions. Both axial and lateral resolution can be improved simul- taneously by virtue of the blinking/fluctuating nature of modified fluorescent probes, exemplified with the quantum dots. Further, super-resolution images with dual modality can be obtained through super-resolution optical fluctuation imaging (SOFI) and bleaching/blinking-assisted localization microscopy (BALM). 4A validated UPLC–MS/MS method for simultaneous determination of imatinib, dasatinib and nilotinib in human plasma显示文摘sensitive, rapid, simple and economical ultra-performance liquid chromatography–tandem mass spectrometric method(UPLC–MS/MS) was developed and validated for simultaneous determination of imatinib,dasatinib and nilotinib in human plasma using gliquidone as internal standard(IS). Liquid-liquid extraction method with ethyl acetate was used for sample pre-treatment. The separation was performed on an Xtimate Phenyl column using isocratic mobile phase consisting of A(aqueous phase: 0.15% formic acid and 0.05% ammonium acetate) and B(organic phase: acetonitrile)(A:B=40:60, v/v). The flow rate was 0.25 mL/min and the total run time was 6 min. The multiple reaction monitoring(MRM) transitions, m/z 494.5→394.5 for imatinib, 488.7→401.5 for dasatinib, 530.7→289.5 for nilotinib and 528.5→403.4 for IS, were chosen to achieve high selectivity in the simultaneous analyses. The method exhibited great improvement in sensitivity and good linearity over the concentration range of 2.6–5250.0 ng/mL for imatinib, 2.0–490.0 ng/mL for dasatinib, and 2.4–4700.0 ng/mL for nilotinib. The method showed acceptable results on sensitivity, specificity,recovery, precision, accuracy and stability tests. This UPLC–MS/MS assay was successfully used for human plasma samples analysis and no significant differences were found in imatinib steady-state trough concentrations among the SLC22A5-1889T > C or SLCO1B3 699G > A genotypes(P > 0.05). 5Key Elements Controlling Oil Accumulation within the Tight Sandstones显示文摘Tight oil sandstone reservoirs with low porosity and permeability, which are an unconventional petroleum resource, have been discovered in the Jurassic intervals of the central Junggar Basin, the northwestern China. To reveal the accumulation mechanism, a relatively comprehensive research was conducted, including oil-source correlation, porosity evolution, and hydrocarbon charging history. The results show that crude oil of these tight sandstone reservoirs were mainly from Permian source rocks with some contribution from Jurassic source rocks. The reservoirs were buried at shallow depth(<3 100 m) and exposed to weak diagenesis, and thus had high porosity(18.5%) when the Permian-sourced oil from Permian source rock was charging, indicating high GOI values(>5%). In contrast, the sandstone reservoir had already become tight and did not provide available space to accumulate oil due to severe compaction and cementation when hydrocarbon from Jurassic source rock filled, evidenced by low GOI values(<5%). Therefore, reservoir porosity controls the oil accumulation within tight sandstone. Whether tight sandstone reservoirs accumulate oil depends on the reservoir quality when hydrocarbons charge. 7Sea ice thickness analyses for the Bohai Sea using MODIS thermal infrared imagery显示文摘Level ice thickness distribution pattern in the Bohai Sea in the winter of 2009–2010 was investigated in this paper using MODIS night-time thermal infrared imagery.The cloud cover in the imagery was masked out manually.Level ice thickness was calculated using MODIS ice surface temperature and an ice surface heat balance equation.Weather forcing data was from the European Centre for Medium-Range Weather Forecasts(ECMWF) analyses.The retrieved ice thickness agreed reasonable well with in situ observations from two off-shore oil platforms.The overall bias and the root mean square error of the MODIS ice thickness are –1.4 cm and 3.9 cm,respectively.The MODIS results under cold conditions(air temperature < –10°C) also agree with the estimated ice growth from Lebedev and Zubov models.The MODIS ice thickness is sensitive to the changes of the sea ice and air temperature,in particular when the sea ice is relatively thin.It is less sensitive to the wind speed.Our method is feasible for the Bohai Sea operational ice thickness analyses during cold freezing seasons.ZENG Tao SHI Lijian MARKO Makynen CHENG Bin ZOU Juhong ZHANG Zhiping 2016Acta Oceanologica Sinica2016,35,7:3
8Changes in secretory pathway Ca^(2+)-ATPase 2 following focal cerebral ischemia/reperfusion injury显示文摘This study aimed to investigate changes in secretory pathway Ca2+-ATPase 2 expression following cerebral ischemia/reperfusion injury, and to define the role of Ca2+-ATPases in oxidative stress. A rat model of cerebral ischemia/reperfusion injury was established using the unilateral middle cerebral artery occlusion method. Immunohistochemistry and reverse transcription-PCR assay results showed that compared with the control group, the expression of secretory pathway Ca2+-ATPase 2 protein and mRNA in the cerebral cortex and hippocampus of male rats did not significantly change during the ischemic period. However, secretory pathway Ca2+-ATPase 2 protein and mRNA expression reduced gradually at 1, 3, and 24 hours during the reperfusion period. 10Cross-cumulant enhanced radiality nanoscopy for multicolor superresolution subcellular imaging显示文摘Fluorescence fuctuation-based superresolution techniques can achieve fast superresolution imaging on a cost-effective wide-field platform at a low light level with reduced phototoxicity.However,the current methods exhibit certain imaging deficiencies that misinterpret nanoscale features reconstructed from fluctuating image scquences,thus degrading the superresolution imaging quality and performance.Here we propose Cross-cumulant enhanced radiality nanoscopy(CERN),which employs cross cumulant analysis in tandem with radiality processing.We demonstrated that CERN can significantly improve the spatial resolution at a low light level while eliminating the misinterpretations of nanoscale features of the existing fluctuation-based superresolution methods.In the experiment,we further verified the superior performance of CERN over the current methods through performing multicolor superresolution imaging of subcellular microtubule networks and clathrin-coated pits as well as the high-precision reconstruction of densely packed RNA transcripts.ZHIPING ZENG JING MA CANHUA Xu 2020Photonics Research2020,8,6:2
11Three-dimensional (3D) structure model and its parameters for poplar shelterbelts显示文摘The spatial functions of surface area density(vegetative surface area per unit canopy volume) and cubic density(vegetative volume per unit canopy volume) have been used as two three-dimensional(3D) structural descriptors for shelterbelt.The functions were defined by models as a general case.However,sub-models such as surface area,volume,and corresponding distributions were not explicitly defined for poplar trees,which are a dominant woody species in shelterbelts all over China,and this limits applications of the models in China and elsewhere.In order to define and develop these sub-models for shelterbelts,poplar trees were destructively sampled from multiple-row shelterbelts and then were measured for their surface area and volume.Using these measurements,we estimated parameters to define their equations explicitly.Based on the architecture and planting patterns of trees in shelterbelts,the distribution of the surface areas and volumes vertically and across the width for different tree heights were constructed for the three components of trunks,branches and leaves.Incorporating the defined equations into the models,we described the 3D structure of a multiple-row poplar shelterbelt.The results showed that,the spatial change in magnitude of surface area density(0.215-10.131 m2/m3) or cubic density(0.00007-0.04667 m3/m3) in shelterbelts is large and their distributions are not uniform.The assumption for boundary-layer flow modeling efforts that the 3D distribution of shelterbelt structure was uniform is not the case in field.The 3D structure model not only can be used to model the flow field as influenced by each tree component,but also can express the entire aerodynamic characteristics of a shelterbelt.The methodologies and equations that are developed in this study can be applied to estimate the 3D structure of a shelterbelt with a design similar to our studied poplar shelterbelts in terms of species composition and planting patterns.The fitted models can be used to describe the 3D aerodynamic structure of field shelterbelts.Furthermore,an improved description of shelterbelt structure has great potential to improve the simulation of boundary layer flows as influenced by that shelterbelt.Such insights can eventually be used to quantify the design of shelterbelt structure and/or adjustment for managing the function of shelterbelts and their effects on microclimate.FAN ZhiPing GAO JunGang ZENG DeHui ZHOU XinHua SUN XueKai 2010Science China Earth Sciences2010,53,10:2
