Design and development of an oral remdesivir derivative W116 against SARS-CoV-2显示文摘Dear Editor,Since the declaration of COVID-19 as a global pandemic on March 11,2020,this pandemic has been circulating for 17 months throughout the world, leading to more than 200 million infections and nearly 4.4 million deaths as of August 15,2021.The pathogen of COVID-19 is a novel coronavirus named SARS-CoV-2,which shares-79%genome sequence identity with SARS-CoV.1 At the early stage of the COVID-19 outbreak,5ARS-CoV-2 caused great panic in the hardest-hit areas due to its high transmissibility and pathogenicity. To fight the COVID-19 crisis, drug repurposing was immediately pursued in order to find potential therapeutics. | Yuanchao Xie Wanchao Yin Yumin Zhang Weijuan Shang Zhen Wang Xiaodong Luan Guanghui Tian Haji A.Aisa Yechun Xu Gengfu Xiao Jia Li Hualiang Jiang Shuyang Zhanq Leike Zhang H.Eric Xu Jingshan Shen | 2021 | Cell Research2021,31,11: | 12
Effects of three needling manipulations of the right-side Zusanli(ST 36) on brain using functional magnetic resonance imaging显示文摘OBJECTIVE: To investigate the effects on the brain using three needling manipulations(twirling, lifting-thrusting, and twirling plus lifting-thrusting)when the right-side Zusanli(ST 36) acupoint was stimulated with needles.METHODS: Seventeen healthy subjects accepted three needling manipulations stimulating the right Zusanli(ST 36) over separate days. Functional magnetic resonance imaging was used to detect changes in the brain during the manipulations, and then the needling sensations were recorded using the MGH acupuncture sensation scale(MASS) after each scan. f MRI data were processed using Statistical Parametric Mapping 8 to analyze the positiveand negative activation in the brain induced by different acupuncture manipulations.RESULTS: The individual needling sensations showed no statistically significant differences among the three manipulations. However, the MASS index showed that lifting-thrusting > twirling plus lifting-thrusting > twirling. Lifting-thrusting activated left premotor cortex, left postcentral, right middle frontal gyrus, left inferior frontal gyrus, right lingual gyrus, left insula, right putamen, bilateral cingulate gyrus and right cerebellum; and deactivated bilateral hippocampus and left caudate.Twirling activated bilateral orbital middle frontal gyrus, left opercular and triangular inferior frontal gyrus, and right middle occipital gyrus; and deactivated bilateral precuneus, right amygdala, left anterior cingulate gyrus, right inferior temporal gyrus, right middle frontal gyrus, right supplementary motor gyrus, and left postcentral. Twirling plus lifting-thrusting activated bilateral postcentral, left inferior occipital gyrus, left insula, left thalamus, left cingulate gyrus, and right putamen; and deactivated right superior frontal gyrus, right superior parietal gyrus, right temporal gyrus, right middle occipital gyrus, right insula, and left lingual gyrus. Pairwise comparisons of the three manipulations showed that signals induced by lifting-thrusting were the strongest, especially in the limbic system, followed by twirling plus lifting-thrusting; twirling alone was the weakest.CONCLUSION: Three methods of needling manipulations similarly activated areas associated with the somatosensory system, vision, cognition, and emotional regulation. This may have significant implications for acupuncture in clinical practice. | Lu Fengyan Gao Junhong Wang Yuanyuan Dai Qiufu Bao Yan Hong Yang Zhang Guolei Xin Juanjuan Zhao Yuxue Wu Shuyang Chen Yuanyuan Li Xiaojiao Fang Jiliang Yu Xiaochun | 2017 | Journal of Traditional Chinese Medicine2017,37,3: | 5

Group VB transition metal dichalcogenides for oxygen reduction reaction and strain-enhanced activity governed by p-orbital electrons of chalcogen显示文摘Developi ng alter native oxyge n reducti on reactio n (ORR) catalysts to replace precious Pt-based metals with abundant materials is the key challe nge of commercial application of fuel cells. Owing to their various compositi ons and tun able electronic properties, transition metal dichalcogenides (TMDs) have the great potential to realize high-efficiency catalysts for ORR. Here, various 3R-phase dichalcogenides of group VB and VIB transition metals (MX2, M = Nb, Ta, Mo, W;X = S, Se, Te) are investigated for ORR catalysts by using density functional theory calculations. The computed over-potentials of group VB TMDs are much less than those of group VIB TMDs. For group VB TMDs, a volcano-type plot of ORR catalytic activity is established on the adsorption energies of *OH, and NbS2 and TaTe2 exhibit best ORR activity with an oveepotential of 0.54 V. To achieve even better activity, strain engineering is performed to tune ORR catalytic activity, and the minimum over-potential of 0.43 V can be realized. We further dem on strate that the shift of p orbital center of surface chalcoge n elements under strain is responsible for tuning the catalytic activity of TMDs. | Shuyang Zhao Ke Wang Xiaolong Zou Lin Gan Hongda Du Chengjun Xu Feiyu Kang Wenhui Duan Jia Li | 2019 | Nano Research2019,12,4: | 4
Influence of rare earth Ce on hot deformation behavior of as-cast Mn18Cr18N high nitrogen austenitic stainless steel显示文摘The hot deformation behavior of Mn18Cr18N and Mn18Cr18N+Ce high nitrogen austenitic stainless steels at 1173-1473 K and 0.01-1 s^(-1) were investigated by thermal compression tests. The influence mechanism of Ce on the hot deformation behavior was analyzed by Ce-containing inclusions and segregation of Ce.The results show that after the addition of Ce,large, angular, hard, and brittle inclusions(TiN-Al_(2)O_(3),TiN,and Al_(2)O_(3)) can be modified to fine and dispersed Ce-containing inclusions(Ce-Al-O-S and TiN-Ce-Al-O-S).During the solidification,Ce-containing inclusions can be used as heterogeneous nucleation particles to refine as-cast grains.During the hot deformation,Ce-containing inclusions can pin dislocation movement and grain boundary migration, induce dynamic recrystallization(DRX)nucleation, and avoid the formation and propagation of micro cracks and gaps.In addition, during the solidification,Ce atoms enrich at the front of solid-li-quid interface, resulting in composition supercooling and refining the secondary dendrites.Similarly, during the hot deformation,Ce atoms tend to segregate at the boundaries of DRX grains, inhibiting the growth of grains.Under the synergistic effect of Ce-containing inclusions and Ce segregation, although the hot deformation resistance and hot deformation activation energy are improved,DRX is more likely to occur and the size of DRX grains is significantly refined, and the problem of hot deformation cracking can be alleviated. Finally, the microhardness of the samples was measured. The results show that compared with as-cast samples, the microhardness of hot-deformed samples increases signific-antly,and with the increase of DRX degree, the microhardness decreases continuously. In addition,Ce can affect the microhardness of Mn18Cr18N steel by affecting as-cast and hot deformation microstructures. | Yushuo Li Yanwu Dong Zhouhua Jiang Qingfei Tang Shuyang Du Zhiwen Hou | 2023 | International Journal of Minerals,Metallurgy and Materials2023,30,2: | 4
Application of next-generation sequencing to screen for pathogenic mutations in 123 unrelated Chinese patients with Marfan syndrome or a related disease显示文摘Marfan syndrome(MFS) is a systemic connective tissue disease principally affecting the ocular, skeletal and cardiovascular systems. This autosomal dominant disorder carries a prevalence of 1:3,000 to 1:5,000. This study aims to define the mutational spectrum of MFS related genes in Chinese patients and to establish genotype-phenotype correlations in MFS. Panel-based targeted next-generation sequencing was used to analyze the FBN1, TGFBR1 and TGFBR2 genes in 123 unrelated Chinese individuals with MFS or a related disease. Genotype-phenotype correlation analyses were performed in mutation-positive patients. The results showed that 97 cases/families(78.9%;97/123) harbor at least one(likely) pathogenic mutation, most of which were in FBN1;four patients had TGFBR1/2 mutations;and one patient harbored a SMAD3 mutation. Three patients had two FBN1 mutations, and all patients showed classical MFS phenotypes. Patients with a dominant negative-FBN1 mutation had a higher prevalence of ectopia lentis(EL). Patients carrying a haploinsufficiency-FBN1 mutation tended to have aortic dissection without EL. This study extends the spectrum of genetic backgrounds of MFS and enriches our knowledge of genotype-phenotype correlations. | Jiacheng Li Chaoxia Lu Wei Wu Yaping Liu Rongrong Wang Nuo Si Xiaolu Meng Shuyang Zhang Xue Zhang | 2019 | Science China(Life Sciences)2019,62,12: | 4

COVID-19-associated monocytic encephalitis(CAME):histological and proteomic evidence from autopsy显示文摘Severe neurological symptoms are associated with Coronavirus disease 2019(COVID-19).However,the morphologic features, pathological nature and their potential mechanisms in patient brains have not been revealed despite evidence of neurotropic infection. In this study, neuropathological damages and infiltrating inflammatory cells were quantitatively evaluated by immunohistochemical staining, ultrastructural examination under electron microscopy, and an image threshold method,in postmortem brains from nine critically ill COVID-19 patients and nine age-matched cadavers of healthy individuals.Differentially expressed proteins were identified by quantitative proteomic assays.Histopathological findings included neurophagocytosis, microglia nodules, satellite phenomena, extensive edema, focal hemorrhage, and infarction,as well as infiltrating mononuclear cells.Immunostaining of COVID-19 brains revealed extensive activation of both microglia and astrocytes, severe damage of the blood-brain barrier(BBB)and various degrees of perivascular infiltration by predominantly CD14+/CD16+/CD141+/CCR7+/CD11c+monocytes and occasionally CD4+/CD8+T lymphocytes.Quantitative proteomic assays combined with bioinformatics analysis identified upregulated proteins predominantly involved in immune responses, autophagy and cellular metabolism in COVID-19 patient brains compared with control brains.Proteins involved in brain development, neuroprotection, and extracellular matrix proteins of the basement membrane were downregulated, potentially caused by the activation of transforming growth factorβreceptor and vascular endothelial growth factor signaling pathways. Thus, our results define histopathological and molecular profiles of COVID-19-associated monocytic encephalitis(CAME)and suggest potential therapeutic targets. | Pei-Pei Zhang Zhi-Cheng He Xiao-Hong Yao Rui Tang Jie Ma Tao Luo Chuhong Zhu Tian-Ran Li Xindong Liu Dingyu Zhang Shuyang Zhang Yi-Fang Ping Ling Leng Xiu-Wu Bian | 2023 | Signal Transduction and Targeted Therapy2023,8,2: | 3
SRSF1 plays a critical role in invariant natural killer T cell development and function显示文摘Invariant natural killer T(iNKT)cells are highly conserved innate-like T lymphocytes that originate from CD4^(+)CD8^(+)double-positive(DP)thymocytes.Here,we report that serine/arginine splicing factor 1(SRSF1)intrinsically regulates iNKT cell development by directly targeting Myb and balancing the abundance of short and long isoforms.Conditional ablation of SRSF1 in DP cells led to a substantially diminished iNKT cell pool due to defects in proliferation, survival, and TCRαrearrangement. The transition from stage 0 to stage 1 of iNKT cells was substantially blocked, and the iNKT2 subset was notably diminished in SRSF1-deficient mice.SRSF1 deficiency resulted in aberrant expression of a series of regulators that are tightly correlated with iNKT cell development and iNKT2 differentiation, including Myb,PLZF,Gata3,ICOS,and CD5.In particular,we found that SRSF1 directly binds and regulates pre-mRNA alternative splicing of Myb and that the expression of the short isoform of Myb is substantially reduced in SRSF1-deficient DP and iNKT cells.Strikingly, ectopic expression of the Myb short isoform partially rectified the defects caused by ablation of SRSF1.Furthermore,we confirmed that the SRSF1-deficient mice exhibited resistance to acute liver injury uponα-GalCer and Con A induction. Our findings thus uncovered a previously unknown role of SRSF1 as an essential post-transcriptional regulator in iNKT cell development and functional differentiation, providing new clinical insights into iNKT-correlated disease. | Jingjing Liu Menghao You Yingpeng Yao Ce Ji Zhao Wang Fang Wang Di Wang Zhihong Qi Guotao Yu Zhen Sun Wenhui Guo Juanjuan Liu Shumin Li Yipeng Jin Tianyan Zhao Hai-Hui Xue Yuanchao Xue Shuyang Yu | 2021 | Cellular & Molecular Immunology2021,18,11: | 3
Exome sequencing reveals genetic architecture in patients with isolated or syndromic short stature显示文摘Short stature is among the most common endocrinological disease phenotypes of childhood and may occur as an isolated finding or in conjunction with other clinical manifestations.Although the diagnostic utility of clinical genetic testing in short stature has been implicated, the genetic architecture and the utility of genomic studies such as exome sequencing(ES)in a sizable cohort of patients with short stature have not been investigated systematically. In this study,we recruited 561 individuals with short stature from two centers in China during a 4-year period. We performed ES for all patients and available parents. All patients were retrospectively divided into two groups:an isolated short stature group(group I,n=257)and an apparently syndromic short stature group(group II,n=304).Causal variants were identified in 135 of 561(24.1%)patients.In group I,29 of 257(11.3%)of the patients were solved by variants in 24 genes. In group II,106 of 304(34.9%)patients were solved by variants in 57 genes.Genes involved in fundamental cellularprocess played an important role in the genetic architecture of syndromic short stature.Distinct genetic architectures and pathophysiological processes underlie isolated and syndromic short stature. | Xin Fan Sen Zhao Chenxi Yu Di Wu Zihui Yan Lijun Fan Yanning Song Yi Wang Chuan Li Yue Ming Baoheng Gui Yuchen Niu Xiaoxin Li Xinzhuang Yang Shiyu Luo Qiang Zhang Xiuli Zhao Hui Pan Mei Li Weibo Xia Guixing Qiu Pengfei Liu Shuyang Zhang Jianguo Zhang Zhihong Wu James R.Lupski Jennifer E.Posey Shaoke Chen Chunxiu Gong Nan Wu | 2021 | Journal of Genetics and Genomics2021,48,5: | 2
Gene duplication drove the loss of awn in sorghum.显示文摘Loss of the awn in some cereals including sorghum is a key transition during cereal domestication or improvement that has facilitated grain harvest and storage. The genetic basis for the loss of awn in sorghum during domestication or improvement remains unknown. Here,we identified a transcription factor gene awn1 encoding an ALOG domain, which is responsible for awn loss during sorghum domestication or improvement.awn1 arose from a gene duplication from chromosome 10 that translocated to chromosome 3,recruiting a new promoter from the neighbouring intergenic region filled with'noncoding DNA',and recreating the first exon and intron. The awn1 acquires high expr`ession after duplication and represses the elongation of awns in domesticated sorghum.Comparative mapping revealed a high collinearity at awn1 paralog locus on chromosome 10 across cereals and awn growth and development was successfully reactivated on the rice spikelet by inactivating rice awn1 orthologue.Further RNA-seq and DAP-seq revealed that as a transcription repressor,AWN1 directly bound to the motif in the regulatory regions from three MADS genes related to flower development and two genes DL and LKS2 for the development of awn, downregulated the expressions of these genes, and then repressed the elongation of awn.The preexistence of regulatory elements in the neighbouring intergenic region of awn1 before domestication signified that noncoding DNA may serve as a treasure trove for evolution during adaptation to a changing world. Our results supported that gene duplication can promptly drive the evolution of gene regulatory network. | Leina Zhou Can Zhu Xiaojian Fang Hangqin Liu Shuyang Zhong Yan Li Jiacheng Liu Yang Song Xing Jian Zhongwei Lin | 2021 | Molecular Plant2021,14,11: | 2

Amelioration of β^(654)-thalassemia in mouse model with the knockdown of aberrantly spliced β-globin mRNA显示文摘Large amounts of aberrantly spliced mRNA from the β654 allele was present in erythroid cells, which might impair the erythropoiesis. A therapeutic strategy for β-thalassemia was explored by knocking down the aberrantly spliced mRNA of β-globin. Lentiviral vector with siRNA fragment targets on the specific portion of β654-globin aberrantly spliced pre-mRNA was constructed. In HeLa β654 cells, the siRNA vector could reduce approximately 60% of aberrantly spliced mRNA, which was assessed by RT-PCR and qRT-PCR. Furthermore, a disease model of β654 thalassemia mice with lentiviral-mediated siRNA was produced by subzonal injection (named Hβi-Hbbth-4/Hbb+ transgenic mice). Our results showed that the hemotological parameters were improved in Hβi-Hbbth-4/Hbb+ transgenic mice. This study provides a potential way for β654-thalassemia therapy by knocking down the aberrantly spliced β-globin mRNA, whilst supporting that the aberrantly spliced β-globin mRNA may aggravate the disease. | Shuyang Xie Wei Li Zhaorui Ren Jingzhi Zhang Xinbin Guo Shu Wang Shuzhen Huang Fanyi Zeng Yi-Tao Zeng | 2008 | Journal of Genetics and Genomics2008,35,10: | 1
Efficient photoreduction strategy for uranium immobilization based on graphite carbon nitride/activated carbon nanocomposites显示文摘Uranium removal from aqueous solutions using environmentally friendly photocatalytic technology is a novel approach for resource recovery.Herein, carbon nitride/activated carbon composite materials(CN/AC)were investigated for U(Ⅵ)reduction under visible light. An exceptional boost in photocatalytic activity was observed for CN/AC composites(up to 70 times over the conventional bulk g-C_(3)N_(4)).The strong interactive conjugatedπ-bond structure between g-C_(3)N_(4) and AC accelerated the migration of carriers and then prolonged the electron lifetime.CN/AC composites exhibited excellent compatibility with different water substrates and were resilience to a wide range of p H changes and abundant competitive anions/cations.Quenching experiments and electron microscopy characterization indicated that U(VI)was reduced by photogenerated electrons and deposited on the edge of CN/AC composites. The low-cost,high-performance carbon-based composite material proposed in this work is a potential candidate for the efficient treatment of radioactive wastewater. | Shuyang Li Zhiwei Niu Duoqiang Pan Zhenpeng Cui Hewen Shang Jie Lian Wangsuo Wu | 2022 | Chinese Chemical Letters2022,33,7: | 1
