3Multiscale patterning of a metallic glass using sacrificial imprint lithography显示文摘Bulk metallic glasses(BMGs)have been developed as a means to achieve durable multiscale,nanotextured surfaces with desirable properties dictated by topography for a multitude of applications.One barrier to this achievement is the lack of a bridging technique between macroscale thermoplastic forming and nanoimprint lithography,which arises from the difficulty and cost of generating controlled nanostructures on complex geometries using conventional top-down approaches.This difficulty is compounded by the necessary destruction of any resulting reentrant structures during rigid demolding.We have developed a generalized method to overcome this limitation by sacrificial template imprinting using zinc oxide(ZnO)nanostructures.It is established that such structures can be grown inexpensively and quickly with tunable morphologies on a wide variety of substrates out of solution,which we exploit to generate the nanoscale portion of the multiscale pattern through this bottom-up approach.In this way,we achieve metallic structures that simultaneously demonstrate features from the macroscale down to the nanoscale,requiring only the top-down fabrication of macro/microstructured molds.Upon detachment of the formed part from the multiscale molds,the ZnO remains embedded in the surface and can be removed by etching in mild conditions to both regenerate the mold and render the surface of the BMGs nanoporous.The ability to pattern metallic surfaces in a single step on length scales from centimeters down to nanometers is a critical step toward fabricating devices with complex shapes that rely on multiscale topography for their intended functions,such as biomedical and electrochemical applications.Jonathan P.Singer Candice I.Pelligra Noga Kornblum Youngwoo Choo Manesh Gopinadhan Punnathat Bordeenithikasem Jittisa Ketkaew Seng Fatt Liew Hui Cao Jan Schroers Chinedum O.Osuji 2015Microsystems & Nanoengineering2015,1,1:2

15The role of porosity and 3D cross-stent configuration of multiple overlapping uncovered stents in the management of complex aortic aneurysms-Insights from haemodynamics显示文摘Background:Multiple overlapping uncovered stents(MOUS)have been introduced to manage complex aortic aneurysms with vital branches involvement.It has been shown that the porosity is a key determinant of the treatment outcome.However,the role of 3D cross-stent configuration remains unclear.Methods:One patient with a complex aortic aneurysm judged not suitable for open surgery nor endovascular repair was invited to participate this study.In total,four bare metal stents were deployed.3D lesion geometry was reconstructed based on pre-and post-operative CTA,and the zero-pressure configuration was recovered using an inverse procedure.Local haemodynamic parameters,including wall shear stress(WSS),oscillatory shear index(OSI),and particle relative resident time(RRT),as well as the vessel structural stress(VSS),were quantified using one-way fluid-structure interaction(FSI)analysis.In comparison to MOUS,a corresponding compact model was reconstructed by projecting inner layer stents to the most outer layer to form a single layer to eliminate the 3D cross-stent configuration and one-way FSI analysis was performed.Results:Results obtained showed that the porosity decreased linearly with the number of stents.When the 1st stent was deployed,the mean velocity decreased 36.4%and further reduction of 49.3%,59.8%,and 62.8%were observed when the 2nd,3rd and 4th stents were deployed.WSS also decreased with the number of stents deployed,and both OSI and RRT increased,but the increase was very minor with the 4th stent.MOUS deployment induced high VSS concentration in the landing zone while the VSS and pressure in the sac remained nearly unchanged.The compact model yielded a small difference in the value of flow-related parameters and 10%–20%reduction in VSS.Conclusion:Compared with porosity,the 3D cross-stent of MOUS configuration plays a minor role in the modulation of local haemodynamics.A compact model does not reduce high VSS concentration in the diseased region significantly.Shuo Wang Yongxue Zhang Jianxuan Feng Yuan Huang Pinjing Hui Jonathan H.Gillard Qingsheng Lu Zhongzhao Teng 2019Medicine in Novel Technology and Devices2019,,3:0
16SRS27,a semisynthetic diterpenoid lactone,inhibits airway inflammation and hyper-responsiveness via NF-κB pathway in an allergic mouse asthma model显示文摘Andrographis paniculata contains two main diterpenoid constituents,andrographolide(AGP)and 14-deoxy-11,12-didehydroandrographolide(DDAG),which were found to exhibit antiasthma effects in a mouse asthma model.However,due to inadequacies of both compounds in terms of drug-likeness,DDAG analogues were semisynthesised to tackle these shortcomings.The objective of the study was to investigate the potential of DDAG analogues as new antiasthma agents.Among the analogues,(SRS27)was proven to inhibit cysteinyl leukotrienes(CysLT)and nitric oxide(NO)synthesis in mouse macrophages,like AGP.DDAG,on the other hand,failed to exhibit such activities.SRS27 is less cytotoxic than AGP,suggests that a simple chemical modification of DDAG produces a compound with CysLT and NO inhibitory activites similar to AGP while maintaining toxicity profile similar to DDAG.It is interesting to note that other analogues such as SRS28,SRS49,SRS76 and SRS83 with chemical modifications on the same carbon numbers 3 and 19 of DDAG were unable to show inhibition of CysLT and NO synthesis.Consequently,the potential anti-inflammatory effect of SRS27 was investigated in ovalbumin(OVA)-induced mouse asthma model.The compound was administered in a prophylactic manner and showed a substantial decrease in mouse asthma model parameters.SRS27 at 3mg·kg-1 significantly reduced OVA-induced total cell such as macrophages,eosinophils,lymphocytes and neutrophils,as well as inflammatory cytokines such as IL-4,IL-5,IL-13 and eotaxin in bronchoalveolar lavage BAL fluid.The compound also suppressed serum IgE production.In addition,SRS27 suppressed mucus hyper-secretion and expression of inflammatory mediators such as TNF-α,MCP-1,Muc5 ac,RANTES,IL-33 and iNOS.SRS27 is the first known DDAG derivative tested positive in mouse asthma model and as such SRS27 could serve as a prototype prophylactic agent.Jonathan Chee Woei LIM Fera Yiqian GOH Sreenivasa Rao SAGINEEDU Audrey Chee Hui YONG Shiran Mohd SIDIK Nordin Haji LAJIS Wai-Shiu Fred WONG Johnson STANSLAS 2015中国药理学与毒理学杂志2015,29,S1:0

18Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND 19Recommendations for improved tropical cyclone formation and position probabilistic Forecast products显示文摘Prediction of the potentially devastating impact of landfalling tropical cyclones(TCs)relies substantially on numerical prediction systems.Due to the limited predictability of TCs and the need to express forecast confidence and possible scenarios,it is vital to exploit the benefits of dynamic ensemble forecasts in operational TC forecasts and warnings.RSMCs,TCWCs,and other forecast centers value probabilistic guidance for TCs,but the International Workshop on Tropical Cyclones(IWTC-9)found that the“pull-through”of probabilistic information to operational warnings using those forecasts is slow.IWTC-9 recommendations led to the formation of the WMO/WWRP Tropical Cyclone-Probabilistic Forecast Products(TC-PFP)project,which is also endorsed as a WMO Seamless GDPFS Pilot Project.The main goal of TC-PFP is to coordinate across forecast centers to help identify best practice guidance for probabilistic TC forecasts.TC-PFP is being implemented in 3 phases:Phase 1(TC formation and position);Phase 2(TC intensity and structure);and Phase 3(TC related rainfall and storm surge).This article provides a summary of Phase 1 and reviews the current state of the science of probabilistic forecasting of TC formation and position.There is considerable variability in the nature and interpretation of forecast products based on ensemble information,making it challenging to transfer knowledge of best practices across forecast centers.Communication among forecast centers regarding the effectiveness of different approaches would be helpful for conveying best practices.Close collaboration with experts experienced in communicating complex probabilistic TC information and sharing of best practices between centers would help to ensure effective decisions can be made based on TC forecasts.Finally,forecast centers need timely access to ensemble information that has consistent,user-friendly ensemble information.Greater consistency across forecast centers in data accessibility,probabilistic forecast products,and warnings and their communication to users will produce more reliable information and support improved outcomes.Jason P.Dunion Chris Davis Helen Titley Helen Greatrex Munehiko Yamaguchi John Methven Raghavendra Ashrit Zhuo Wang Hui Yu Anne-Claire Fontan Alan Brammer Matthew Kucas Matthew Ford Philippe Papin Fernando Prates Carla Mooney Andrew Kruczkiewicz Paromita Chakraborty Andrew Burton Mark DeMaria Ryan Torn Jonathan L.Vigh 2023Tropical Cyclone Research and Review2023,12,4:0
20The efficacies and biomarker investigations of antiprogrammed death-1 (anti-PD-1)-based therapies for metastatic bone and soft tissue sarcoma显示文摘Objective:Sarcomas are a group of rare malignancies with various subtypes.Patients with metastatic sarcoma who have failed traditional treatments can possibly achieve better prognoses from using novel therapies,including anti-programmed death-1(PD-1)-based therapies.Methods:We retrospectively analyzed clinical data of 24 metastatic sarcoma patients from June 15,2016 to December 30,2019.These patients mainly received angiogenesis inhibitors combined with anti-PD-1 therapy after they became resistant to traditional treatments.Furthermore,8 patients underwent panel DNA and whole transcript sequencing.Results:Six patients received 2 cycles of anti-PD-1 therapy and were included in the safety evaluation only group.The median follow-up time was 5.77 months.The median progression-free survival was 7.59 months,the overall response rate was 16.7%and the disease control rate was 55.6%.Based on whole exome and transcript sequencing data,there was no association between TMB,TNB,MSI,HLA-LOH,and PD-L1 expressions and sarcoma types with clinical responses.Immunotherapy efficacy and bioinformatics analyses indicated higher intratumoral heterogeneity(ITH)in progressive disease(PD)patients and lower ITH in partial response(PR)and stable disease patients.A higher percentage of immune cell infiltration,especially monocytes,was observed in PR patients.Active stromal gene expression was increased in PD patients but decreased in PR patients.Enrichment analysis revealed that an increased TGF-βsignaling pathway was reversely correlated with anti-PD-1 efficacy,while a decreased inflammatory response signaling pathway was positively correlated with anti-PD-1 efficacy.Conclusions:Our study showed PD-1 inhibitors combined with anti-angiogenesis agents were effective and well-tolerated.ITH,monocyte ratio,stroma subtypes,and the status of immune-associated signaling pathways may be related with anti-PD-1 based therapy.Jia Lu Ting Li Zhichao Liao Hui Yu Yongtian Zhao Haixiao Wu Zhiwu Ren Jun Zhao Ruwei Xing Sheng Teng Yun Yang Xiangchun Li Kexin Chen Jonathan Trent Jilong Yang 2022Cancer Biology & Medicine2022,19,6:0
