3Selection and characterization of the novel anti-human PD-1 FV78 antibody from a targeted epitope mammalian cell-displayed antibody library显示文摘Currently,display-based methods are well established and widely used in antibody engineering for affinity maturation and structural stability improvement.We obtained a novel anti-human programmed death 1(PD-1)antibody using computer-aided design and a mammalian cell display technology platform.We used computer-aided modeling and distance geometry methods to predict and assign the key residues that contributed to the binding of human PD-L1 to PD-1.Then,we analyzed the sequence of nivolumab(an anti-human PD-1 antibody,referred to as MIL75 in the article)to determine the template for antibody design and library construction.We identified a series of potential substitutions on the obtained template and constructed a virtual epitope-targeted antibody library based on the physicochemical properties and each possible location of the assigned key residues.The virtual antibody libraries were displayed on the surface of mammalian cells as the antigen-binding fragments of full-length immunoglobulin G.Then,we used flow cytometry and sequencing approaches to sort and screen the candidates.Finally,we obtained a novel anti-human PD-1 antibody named FV78.FV78 competitively recognized the PD-1 epitopes that interacted with MIL75 and possessed an affinity comparable to MIL75.Our results implied that FV78 possessed equivalent bioactivity in vitro and in vivo compared with MIL75,which highlighted the probability and prospect of FV78 becoming a new potential antibody therapy.Longlong Luo Shi Wang Xiaoling Lang Tingting Zhou Jing Geng Xinying Li Chunxia Qiao Jiannan Feng Beifen Shen Ming Lv Yan Li 2018Cellular & Molecular Immunology2018,15,2:4
4Acrolein Aggravates Secondary Brain Injury After Intracerebral Hemorrhage Through Drp1-Mediated Mitochondrial Oxidative Damage in Mice显示文摘Clinical advances in the treatment of intracranial hemorrhage(ICH)are restricted by the incomplete understanding of the molecular mechanisms contributing to secondary brain injury.Acrolein is a highly active unsaturated aldehyde which has been implicated in many nervous system diseases.Our results indicated a significant increase in the level of acrolein after ICH in mouse brain.In primary neurons,acrolein induced an increase in mitochondrial fragmentation,loss of mitochondrial membrane potential,generation of reactive oxidative species,and release of mitochondrial cytochrome c.Mechanistically,acrolein facilitated the translocation of dynaminrelated protein 1(Drpl)from the cytoplasm onto the mitochondrial membrane and led to excessive mitochondrial fission.Further studies found that treatment with hydralazine(an acrolein scavenger)significantly reversed Drpl translocation and the morphological damage of mitochondria after ICH.In parallel,the neural apoptosis,brain edema,and neurological functional deficits induced by ICH were also remarkably alleviated.In conclusion,our results identify acrolein as an important contributor to the secondary brain injury following ICH.Meanwhile,we uncovered a novel mechanism by which Drpl-mediated mitochondrial oxidative damage is involved in acroleininduced brain injury.Xun Wu Wenxing Cui Wei Guo Haixiao Liu Jianing Luo Lei Zhao Hao Guo Longlong Zheng Hao Bai Dayun Feng Yan Qu 2020Neuroscience Bulletin2020,36,10:3
5A low-firing melilite ceramic Ba2CuGe207 and compositional modulation on microwave dielectric properties through Mg substitution显示文摘A melilite Ba2CuGe2O7 ceramic was characterized by low sintering temperature and moderate microwave dielectric properties.Sintered at 960℃,the Ba2CuGe2O7 ceramic had a high relative density 97%,a low relative permittivity(εr)9.43,a quality factor(Q×f)of 20,000 GHz,and a temperature coefficient of resonance frequency(τf)-76 ppm/℃.To get a deep understanding of the relationship between composition,structure,and dielectric performances,magnesium substitution for copper in Ba2CuGe2O7 was conducted.Influences of magnesium doping on the sintering behavior,crystal structure,and microwave dielectric properties were studied.Mg doping in Ba2CuGe2O7 caused negligible changes in the macroscopic crystal structure,grain morphology,and size distribution,while induced visible variation in the local structure as revealed by Raman analysis.Microwave dielectric properties exhibit a remarkable dependence on composition.On increasing the magnesium content,the relative permittivity featured a continuous decrease,while both the quality factor and the temperature coefficient of resonance frequency increased monotonously.Such variations in dielectric performances were clarified in terms of the polarizability,packing fraction,and band valence theory.Changzhi YIN Zezong YU Longlong SHU Laijun LIU Yan CHEN Chunchun LI 2021Journal of Advanced Ceramics2021,10,1:3

11Seryl tRNA synthetase cooperates with POT1 to regulate telomere length and cellular senescence显示文摘Deregulated telomere length is a causative factor in many physiological and pathological processes,including aging and cancer.Many studies focusing on telomeres have revealed important roles for cooperation between the Shelterin protein complex and telomerase in maintaining telomere length.However,it remains largely unknown whether and how aging-related stresses,such as deregulated protein homeostasis,impact telomere length.Here,we explored the possible roles of aminoacyl tRNA synthetases(AARSs),key enzymes catalyzing the first reactions in protein synthesis,in regulating telomere length and aging.We selected seryl tRNA synthetase(SerRS)since our previous studies discovered expanded functions of SerRS in the nucleus in addition to its canonical cytoplasmic role in protein synthesis.In this study,we revealed that overexpression of SerRS promoted cellular senescence and inhibited the growth of cervical tumor xenografts in mice by triggering the senescence of tumor cells.In the nucleus,SerRS directly bound to telomeric DNA repeats and tethered more POT1 proteins to telomeres through a direct interaction between the UNE-S domain of SerRS and the OB1 domain of POT1.We further demonstrated that SerRS-induced enrichment of POT1 prevented the recruitment of telomerase to telomeres,resulting in progressive telomere shortening.Our data suggested a possible molecular link between protein synthesis and telomere length control,the deregulation of which may be associated with aging and cancer.Yingxi Li Xiyang Li Mei Cao Yuke Jiang Jie Yan Ze Liu Rongcun Yang Xu Chen Peiqing Sun Rong Xiang Longlong Wang Yi Shi 2019Signal Transduction and Targeted Therapy2019,4,1:1
12TiO^(2)electron transport bilayer for all-inorganic perovskite photodetectors with remarkably improved UV stability toward imaging applications显示文摘High ultraviolet(UV)stability and low dark current(Idark)are necessary for high-quality perovskite photodetectors(PDs).TiO^(2)thin film is known as effective electron-transport-layer(ETL)for perovskite devices.However,common spin-coated TiO^(2)ETLs endow many surface defects and have strong UV photocatalytic effect to decompose perovskite materials,resulting in inferior stability of devices.In this work,TiO^(2)bilayer film(Bi-TiO^(2))has been fabricated by combining spin-coating and atomic-layer-deposition process and its positive effects on UV stability and Idarkof Cs2 AgBiBr6-based PDs have been revealed for the first time.It is demonstrated that Bi-TiO^(2)possesses fewer surface defects and smoother morphology with type II band alignment,which is beneficial to suppress photocatalytic activity of TiO^(2)and reduce carrier recombination at the interface.After accelerated strong UV aging treatment,the PD with Bi-TiO^(2)maintains excellent performance,whereas the PD with spin-coated TiO^(2)film dramatically deteriorate with on-off ratio drops from~102 to~2.Besides,the Idarkof PD remarkably decreases from~10^(-8) A to~10^(-10) A after bilayer optimization.Furthermore,we have integrated the corresponding PDs into a self-built imaging system adopting diffuse reflection mode.This work suggests a feasible approach to fabricate TiO^(2)/Cs2 AgBiBr6-based PDs with remarkable UV tolerance for imaging applications.Ye Yuan Zhong Jia Genghua Yan Zhuowei Li Jinliang Li Min Kuang Bangqi Jiang Longlong Zeng Likun Pan Wenjie Mai 2021Journal of Materials Science & Technology2021,,16:1
13Edge sulfurized graphene nanoplatelets via vacuum mechano-chemical reaction for lithium–sulfur batteries显示文摘Lithium–sulfur batteries have great potential for high energy applications due to their high capacities,low cost and eco-friendliness. However, the particularly rapid capacity decay owing to the dissolution and diffusion of polysulfide intermediate into the electrolyte still hamper their practical applications.And the reported preparation procedures to sulfur based cathode materials are often complex, and hence are rather difficult to produce at large scale. Here, we report a simple mechano-chemical sulfurization methodology in vacuum environment applying ball-milling method combined both the chemical and physical interaction for the one-pot synthesis of edge-sulfurized grapheme nanoplatelets with 3D porous foam structure as cathode materials. The optimal sample of 70%S–Gn Ps-48 h(ball-milled 48 h) obtains 13.2 wt% sulfur that chemically bonded onto the edge of Gn Ps. And the assembled batteries exhibit high initial discharge capacities of 1089 mAh/g at 0.1 C and 950 mAh/g at 0.5 C, and retain a stable discharge capacity of 776 mAh/g after 250 cycles at 0.5 C with a high Coulombic efficiency of over 98%. The excellent performance is mainly attributed to the mechano-chemical interaction between sulfur and grapheme nanoplatelets. 14Biodegradation of occluded hydrocarbons and kerogen macromolecules of the Permian Lucaogou shales,Junggar Basin,NW China显示文摘Three kerogen samples(JJZG-1,JJZG-2 and JJZG-3)isolated from the Permian Lucaogou shales of varying biodegradation levels(BLs≈0,3 and 7,respectively)were subjected to sequential stepwise pyrolysis combined with on-line detection of gas chromatography-mass spectrometry(GC-MS).Occluded fractions(bitumenⅡ)released at low-temperature steps(≥410℃)show consistent biodegradative signatures with that reported for solvent-extracted fractions(bitumenⅠ)of the original shales,e.g.,broad range of abundant n-alkanes,isoprenoids and regular hopanes for the non-biodegraded JJZG-1;trace n-alkanes and abundant hopanes for the moderately biodegraded JJZG-2;and no n-alkanes but still prominent hopanes including the microbially produced 25-nohopanes for the severely biodegraded JJZG-3.This consistency between bitumenⅡand bitumenⅠfractions indicates the biodegradability of the kerogenoccluded bitumenⅡwith limited protection from host kerogen.A minor level of protection was suggested by the trace distribution of n-alkanes in the bitumenⅡof JJZG-2,whereas the bitumenⅠhad no nalkanes.The kerogen itself was more resistant to biodegradation as reflected by the persistence of high abundances of both n-alkanes and hopanes in the high temperature(≥460℃)products of all three kerogen samples.However,the relative abundances of these product groups did show some evidence of biodegradation alteration,e.g.,ratios of n-C_(15)alkene/C_(27)hop-17(21)-ene at 510℃pyrolysis decreased by order of magnitude from the non-biodegraded(JJZG-1=27.4)to highly biodegraded(0.3 for JJZG-3)samples.The reduced biodegradation impact on the kerogen fraction(Cf.bitumen fractions)was also evident by the absence of 25-norhopanes in the high-temperature analysis of the JJZG-3 kerogen.Yan'e Chen Longlong Li Zhirong Zhang Paul F.Greenwood Yumin Liu 2023Energy Geoscience2023,4,1:0
15Surface protection method for the magnetic core using covalent organic framework shells and its application in As(Ⅲ)depth removal from acid wastewater显示文摘Fe_(3)O_(4)-based materials are widely used for magnetic separation from wastewater.However,they often suffer from Fe-leaching behavior under acidic conditions,decreasing their ac-tivity and limiting sustainable practical applications.In this study,covalent organic frame-works(COFs)were used as the shell to protect the Fe_(3)O_(4) core,and the Fe_(3)O_(4)@COF core-shell composites were synthesized for As(Ⅲ)removal from acid wastewater.The imine-linked COFs can in situ grow on the surface of the Fe_(3)O_(4) core layer by layer with[COFs/Fe_(3)O_(4)]mol ratio of up to 2∶1.The Fe-leaching behavior was weakened over a wide pH range of 1-13.Moreover,such composites keep their magnetic characteristic,making them favorable for nanomaterial separation.As(Ⅲ)batch adsorption experiments results indicated that,when COFs are used as the shell for the Fe_(3)O_(4) core,a balance between As(Ⅲ)removal efficiencies and the thickness of the COF shell exists.Higher As(Ⅲ)removal efficiencies are obtained when the[COFs/Fe_(3)O_(4)]mol ratios were<1.5∶1,but thicker COF shells were not beneficial for As(Ⅲ)removal.Such composites also exhibited better As(Ⅲ)removal performances in the pH range of 1-7.Over a wide pH range,the zeta potential of Fe_(3)O_(4)@COF core-shell compos-ites becomes more positive,which benefits the capture of negative arsenic ions.In addition,thinner surface COFs were favorable for mass transfer and facilitating the reaction of Fe and As elements.Our study highlights the promise of using COFs in nanomaterial surface protection and achieving As(Ⅲ)depth removal under acidic conditions.Wenjun Huang Haomiao Xu Xiaoshuang Liu Longlong Wang Shutang Li Leipeng Ji Zan Qu Naiqiang Yan 2022Journal of Environmental Sciences2022,34,5:0

17IMPROVING NITROGEN SAFETY IN CHINA: NITROGEN FLOWS, POLLUTION AND CONTROL显示文摘The impacts of nitrogen on environmental quality,greenhouse gas balances,ecosystem and biodiversity in China are of great concern given the magnitude of demand for food and energy.Comprehensive summaries of historic N flows and their critical threats and sustainable management are urgently needed.This paper initially reviews the historical trends of N flows in China and identifies the critical threats of N loss.Subsequently,it describes some recent success stories of N management,and finally indicates barriers to N pollution control.This review highlights three key points.Firstly,a steady increase of N input in China has led to a series of environmental problems via leaching and runoff,ammonia emissions and denitrification.Secondly,although great efforts to improve N management and N safety in China,further quantifications of N flows and analysis of their underlying mechanisms are needed to improve the understanding of the N cycle and pollution control.Finally,it proposes that the best available technologies combined with regulatory plans,laws,projects and policies should be implemented to overcome current barriers in N control and achieve a balance between the sustainable use of N resources and environmental conservation in China.Chaopu TI Xiaoyuan YAN Longlong XIA Jingwen HUANG 2022Frontiers of Agricultural Science and Engineering2022,9,3:0
18Petrophysical properties and their influencing factors of carbonates in the fourth member of Sinian Dengying Formation, Sichuan Basin, SW China显示文摘Based on the measurements of petrological, petrophysical and elastic properties of the samples of different sedimentary facies in the fourth member of Sinian Dengying Formation (Deng 4 Member) in the Sichuan Basin, the diagenetic processes of reservoirs of different sedimentary facies and their controls on the petrophysical properties were discussed. The results show that cracks and mineral composition jointly control the petrophysical properties, and both are significantly influenced by sedimentary environment and diagenesis. The microbial dolomite of mound-shoal facies mainly experienced multi-stage dolomitization, penecontemporaneous dissolution, tectonic rupture and hydrothermal/organic acid dissolution processes, giving rise to cracks and dissolved pores. The grannular dolomite of inter-mound-shoal bottomland or dolomitic lagoon facies mainly underwent mechanical compaction, burial dolomitization and tectonic-hydrothermal action, creating cracks and intercrystalline pores. The diagenesis related to crack development increases the pressure- and saturation-dependent effects of samples, leading to significant decrease in the compressional wave impedance and Poisson's ratio. Dolomitization changes the properties of mineral particles, resulting in a Poisson's ratio close to dolomite. The muddy, siliceous and calcareous sediments in the low-energy environment lead to the decrease of impedance and the differential change of Poisson's ratio (significantly increased or decreased). 19E6-induced selective translation of WNT4 and JIP2 promotes the progression of cervical cancer via a noncanonical WNT signaling pathway显示文摘mRNA translation reprogramming occurs frequently in many pathologies,including cancer and viral infection.It remains largely unknown whether viral-induced alterations in mRNA translation contribute to carcinogenesis.Most cervical cancer is caused by high-risk human papillomavirus infection,resulting in the malignant transformation of normal epithelial cells mainly via viral E6 and E7 oncoproteins.Here,we utilized polysome profiling and deep RNA sequencing to systematically evaluate E6-regulated mRNA translation in HPV18-infected cervical cancer cells.We found that silencing E6 can cause over a two-fold change in the translation efficiency of~653 mRNAs,most likely in an eIF4E-and eIF2α-independent manner.In addition,we identified that E6 can selectively upregulate the translation of WNT4,JIP1,and JIP2,resulting in the activation of the noncanonical WNT/PCP/JNK pathway to promote cell proliferation in vitro and tumor growth in vivo.Ectopic expression of WNT4/JIP2 can effectively rescue the decreased cell proliferation caused by E6 silencing,strongly suggesting that the WNT4/JIP2 pathway mediates the role of E6 in promoting cell proliferation.Thus,our results revealed a novel oncogenic mechanism of E6 via regulating the translation of mRNAs.Lin Zhao Longlong Wang Chenglan Zhang Ze Liu Yongjun Piao Jie Yan Rong Xiang Yuanqing Yao Yi Shi 2019Signal Transduction and Targeted Therapy2019,4,1:0
20Public key based bidirectional shadow image authentication without pixel expansion in image secret sharing显示文摘Image secret sharing(ISS)is gaining popularity due to the importance of digital images and its wide application to cloud-based distributed storage and multiparty secure computing.Shadow image authentication generally includes shadow image detection and identification,and plays an important role in ISS.However,traditional dealer-participatory methods,which suffer from significant pixel expansion or storing auxiliary information,authenticate the shadow image mainly during the decoding phase,also known as unidirectional authentication.The authentication of the shadow image in the distributing(encoding)phase is also important for the participant.In this study,we introduce a public key based bidirectional shadow image authentication method in ISS without pixel expansion for a(k,n)threshold.When the dealer distributes each shadow image to a corresponding participant,the participant can authenticate the received shadow image with his/her private key.In the decoding phase,the dealer can authenticate each received shadow image with a secret key;in addition,the dealer can losslessly decode the secret image with any k or more shadow images.The proposed method is validated using theoretical analyses,illustrations,and comparisons.Xuehu YAN Longlong LI Jia CHEN Lei SUN 2023Frontiers of Information Technology & Electronic Engineering2023,24,1:0
