Dietary resveratrol improves antioxidant status of sows and piglets and regulates antioxidant gene expression in placenta by Keap1-Nrf2 pathway and Sirt1显示文摘Background: Resveratrol, a plant phenol, affords protection against inflammation and oxidative stress. The objective of this study was to investigate the effects of dietary resveratrol supplementation during pregnancy and lactation on the antioxidant status of sows and piglets and on antioxidant gene expression and pathway in placenta. Methods: Forty sows were allotted to 2 dietary treatments 20 d after breeding. Sows were fed a control diet and a control diet with 300 mg/kg resveratrol. Oxidative stress biomarkers and antioxidant enzymes were measured in the placenta, milk, and plasma of sows and piglets. Antioxidant gene expression and protein expression of Kelch-like ECH-associated protein 1-Nuclear factor E2-related factor 2(Keap1-Nrf2), nuclear factor kappa B-p65(NFκB-p65) and sirtuin1(Sirt1) were quantified in the placenta. Results: Dietary resveratrol increased the litter and piglets weaning weights. Antioxidant status in the milk, placenta and plasma of sows and piglets was partially improved by dietary resveratrol. In placenta, Nrf2 protein expression was increased and Keap1 protein expression was decreased by dietary resveratrol. The m RNA expression of antioxidant genes including catalase(CAT), glutathione peroxidase 1(GPX1), GPX4, superoxide dismutase 1(SOD1)and heme oxygenase 1(HO1), and phase 2 detoxification genes, including glutamate-cysteine ligase modifier(GCLM), microsomal glutathione S-transferase 1(MGST1) and UDP glucuronosyltransferase family 1 member A1(UGT1 A1), was increased by dietary resveratrol. Dietary resveratrol also increased Sirt1 and phosphorylated NFκB-p65 protein expression in the placenta. We failed to observe any influences of dietary resveratrol on pro-inflammatory cytokine levels, including those of interleukin 1β(IL-1β), IL-6, IL-8 and tumor necrosis factor α(TNF-α). However, we observed that the m RNA expression of IL-8 in placenta was reduced by maternal resveratrol. In addition, dietary resveratrol showed interactive effects with day of lactation on activities of SOD and CAT and levels of malonaldehyde(MDA) and hydrogen peroxide(H2 O2) in milk. Conclusions: Dietary resveratrol supplementation during pregnancy and lactation improves the antioxidant status of sows and piglets, which is beneficial to the reproductive performance of sows. Dietary resveratrol regulates placental antioxidant gene expression by the Keap1-Nrf2 pathway and Sirt1 in placenta. | Qingwei Meng Tao Guo Gaoqiang Li Shishuai Sun Shiqi He Baojing Cheng Baoming Shi Anshan Shan | 2018 | Journal of Animal Science and Biotechnology2018,9,3: | 33

Proteomics provides individualized options of precision medicine for patients with gastric cancer显示文摘While precision medicine driven by genome sequencing has revolutionized cancer care, such as lung cancer, its impact on gastric cancer(GC)has been minimal.GC patients are routinely treated with chemotherapy, but only a fraction of them receive the clinical benefit. There is an urgent need to develop biomarkers or algorithms to select chemo-sensitive patients or apply targeted therapy. Here,we carried out retrospective analyses of 1,020 formalin-fixed,paraffin-embedded GC surgical resection samples from 5 hospitals and developed a mass spectrometry-based workflow for proteomic subtyping of GC.We identified two proteomic subtypes:the chemo-sensitive group(CSG)and the chemo-insensitive group(CIG)in the discovery set.The 5-year overall survival of CSG was significantly improved in patients who had received adjuvant chemotherapy after surgery compared with those who received surgery only(64.2%vs.49.6%;Cox P-value=0.002),whereas no such improvement was observed in CIG(50.0%vs.58.6%;Cox P-value=0.495).We validated these results in an independent validation set.Further, differential proteome analysis uncovered 9 FDA-approved drugs that may be applicable for targeted therapy of GC.A prospective study is warranted to test these findings for future GC patient care. | Wenwen Huang Dongdong Zhan Yazhuo Li Nairen Zheng Xin Wei Bin Bai Kecheng Zhang Mingwei Liu Xuefei Zhao Xiaotian Ni Xia Xia Jinwen Shi Cheng Zhang Zhihao Lu Jiafu Ji Juan Wang Shiqi Wang Gang Ji Jipeng Li Yongzhan Nie Wenquan Liang Xiaosong Wu Jianxin Cui Yongsheng Meng Feilin Cao Tieliu Shi Weimin Zhu Yi Wang Lin Chen Qingchuan Zhao Hongwei Wang Lin Shen Jun Qin | 2021 | Science China(Life Sciences)2021,64,8: | 3
Cortical and Subcortical Grey Matter Abnormalities in White Matter Hyperintensities and Subsequent Cognitive Impairment显示文摘Grey matter(GM)alterations may contribute to cognitive decline in individuals with white matter hyperintensities(WMH)but no consensus has yet emerged. Here,we investigated cortical thickness and grey matter volume in 23 WMH patients with mild cognitive impairment(WMH-MCI),43 WMH patients without cognitive impairment, and 55 healthy controls. Both WMH groups showed GM atrophy in the bilateral thalamus,fronto-insular cortices, and several parietal-temporal regions, and the WMH-MCI group showed more extensive and severe GM atrophy. The GM atrophy in the thalamus and fronto-insular cortices was associated with cognitive decline in the WMH-MCI patients and may mediate the relationship between WMH and cognition in WMH patients.Furthermore, the main results were well replicated in an independent dataset from the Alzheimer's Disease Neuroimaging Initiative database and in other control analyses. These comprehensive results provide robust evidence of specific GM alterations underlying WMH and subsequent cognitive impairment. | Wenhao Zhu Hao Huang Shiqi Yang Xiang Luo Wenzhen Zhu Shabei Xu Qi Meng Chengchao Zuo Yong Liu Wei Wang Alzheimer’s Disease Neuroimaging Initiative | 2021 | Neuroscience Bulletin2021,37,6: | 1
Unique and complementary suppression of cGAS-STING and RNA sensing- triggered innate immune responses by SARS-CoV-2 proteins显示文摘The emergence of SARS-CoV-2 has resulted in the COVID-19 pandemic, leading to millions of infections and hundreds of thousands of human deaths. The efficient replication and population spread of SARS-CoV-2 indicates an effective evasion of human innate immune responses, although the viral proteins responsible for this immune evasion are not clear. In this study,we identified SARS-CoV-2 structural proteins, accessory proteins, and the main viral protease as potent inhibitors of host innate immune responses of distinct pathways. In particular, the main viral protease was a potent inhibitor of both the RLR and cGAS-STING pathways.Viral accessory protein 0RF3a had the unique ability to inhibit STING,but not the RLR response. On the other hand, structural protein N was a unique RLR inhibitor.0RF3a bound STING in a unique fashion and blocked the nuclear accumulation of p65 to inhibit nuclear factor-KB signaling.3CL of SARS-CoV-2 inhibited K63-ubiquitin modification of STING to disrupt the assembly of the STING functional complex and downstream signaling.Diverse vertebrate STINGs,including those from humans, mice, and chickens, could be inhibited by 0RF3a and 3CL of SARS-CoV-2.The existence of more effective innate immune suppressors in pathogenic coronaviruses may allow them to replicate more efficiently in vivo.Since evasion of host innate immune responses is essential for the survival of all viruses, our study provides insights into the design of therapeutic agents against SARS-CoV-2. | Yajuan Rui Jiaming Su Si Shen Ying Hu Dingbo Huang Wenwen Zheng Meng Lou Yifei Shi Meng Wang Shiqi Chen Na Zhao Qi Dong Yong Cai Rongzhen Xu Shu Zheng Xiao-Fang Yu | 2021 | Signal Transduction and Targeted Therapy2021,6,4: | 1

Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector. Some of JUNO's features make it an excellent location for^8B solar neutrino measurements, such as its low-energy threshold, high energy resolution compared with water Cherenkov detectors, and much larger target mass compared with previous liquid scintillator detectors. In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable, assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition, approximately 60,000 signal and 30,000 background events are expected. This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter, which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle. Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better, depending on the central value, and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND experiment. | 10 | Stable multi-electron reaction stimulated by W doping VS_(4)for enhancing magnesium storage performance显示文摘Rechargeable magnesium batteries(RMBs)hold promise for offering higher volumetric energy density and safety features, attracting increasing research interest as the next post lithium-ion batteries.Developing high performance cathode material by inducing multi-electron reaction process as well as maintaining structural stability is the key to the development and application of RMBs.Herein, multielectron reaction occurred in VS_(4)by simple W doping strategy.W doping induces valence of partial V as V^(2+)and V^(3+)in VS_(4)structure,and then stimulates electrochemical reaction involving multi-electrons in 0.5%W-V-S.The flower-like microsphere morphology as well as rich S vacancies is also modulated by W doping to neutralize structure change in such multi-electron reaction process. The fabricated 0.5%W-V-S delivers higher specific capacity(149.3 m A h g^(-1)at 50 m A g^(-1),which is 1.6 times higher than that of VS_(4)),superior rate capability(76 mA h g^(-1)at 1000 mA g^(-1)),and stable cycling performance(1500cycles with capacity retention ratio of 93.8%).Besides that,pesudocapaticance-like contribution analysis as well as galvanostatic intermittent titration technique(GITT)further confirms the enhanced Mg^(2+)storage kinetics during such multi-electron involved electrochemical reaction process.Such discovery provides new insights into the designing of multi-electron reaction process in cathode as well as neutralizing structural change during such reaction for realizing superior electrochemical performance in energy storage devices. | Yuxin Tian Jiankang Chen Guofeng Wang Bing Sun Alan Meng Lei Wang Guicun Li Jianfeng Huang Shiqi Ding Zhenjiang Li | 2024 | Journal of Energy Chemistry2024,89,2: | 0

Multidimensional defects tailoring local electron and Mg^(2+) diffusion channels for boosting magnesium storage performance of WO_(3)/MoO_(2)显示文摘Defect engineering presents great promise in addressing lower specific capacity, sluggish diffusion kinetics and poor cycling life issues in energy storage devices.Herein,multidimensional(0D/2D/3D) structural defects are constructed in WO_(3)/MoO_(2) simultaneously via competing for and sharing with O atoms during simple hydrothermal process.OD and 2D defects tailor local electron, activating more sites and generating built-in electric fields to yield ion reservoir,meanwhile,3D defect owning lower anisotropic property tailors Mg^(2+) diffusion channels to fully exploit Mg^(2+) adsorbed sites induced by OD and 2D defects, enhance the kinetics and maintain structural stability.Benefitted from synergistic effect of 0D/2D/3D structural defects, the designed WO_(3)/MoO_(2) shows the higher specific capacity(112.8 mA h g^(-1) at 50 mA g^(-1) with average attenuation rate per cycle of 0.068%),superior rate capability and excellent cycling stability(specific capacity retention of 80% after 1500 cycles at 1000 mA g^(-1)).This strategy provides design ideas of introducing multidimensional structural defects for tailoring local electron and microstructure to improve energy storage property. | Shiqi Ding Yuxin Tian Jiankang Chen He Lv Amin Wang Jingjie Dai Xin Dai Lei Wang Guicun Li Alan Meng Zhenjiang Li | 2023 | Journal of Energy Chemistry2023,,9: | 0
Atomic insights of electronic states engineering of GaN nanowires by Cu cation substitution for highly efficient lithium ion battery显示文摘Electronic engineering of gallium nitride(Ga N) is critical for enhancement of its electrode performance. In this work, copper(Cu) cation substituted Ga N(Cu-Ga N) nanowires were fabricated to understand the electronically engineered electrochemical performance for Li ion storage. Cu cation substitution was revealed at atomic level by combination of X-ray photoelectron spectroscopy(XPS), X-ray absorption fine structure(XAFS), density functional theory(DFT) simulation, and so forth. The Cu-Ga N electrode delivered high capacity of 813.2 m A h g^(-1) at 0.1 A g^(-1) after 200 cycles, increased by 66% relative to the unsubstituted Ga N electrode. After 2000 cycles at 10 A g^(-1),the reversible capacity was still maintained at326.7 m A h g^(-1). The DFT calculations revealed that Cu substitution introduced the impurity electronic states and efficient interatomic electron migration, which can enhance the charge transfer efficiency and reduce the Li ion adsorption energy on the Cu-Ga N electrode. The ex-situ SEM, TEM, HRTEM, and SAED analyses demonstrated the reversible intercalation Li ion storage mechanism and good structural stability. The concept of atomic-arrangement-assisted electronic engineering strategy is anticipated to open up opportunities for advanced energy storage applications. | Zhenjiang Li Kesheng Gao Ying Han Shiqi Ding Yanglansen Cui Minmin Hu Jian Zhao Meng Zhang Alan Meng Jimmy Yun Zhiming Liu Da-Wei Wang Changlong Sun | 2022 | Journal of Energy Chemistry2022,31,4: | 0

Measurement of CO_(2) adsorption capacity with respect to different pressure and temperature in sub-bituminous: implication for CO_(2) geological sequestration显示文摘CCUS (carbon capture, utilization, and storage) technology is regarded as a bottom method to achieve carbon neutrality globally. CO_(2) storage in deep coal reservoirs serves as a feasible selection for CCUS, and its storage potential can be attributed to the CO_(2) adsorption capacity of the coal. In this paper, a series of CO_(2) adsorption isotherm experiments were performed at different pressures and temperatures in sub-bituminous coal from the southern Junggar Basin (reservoir temperature ∼25.9°C and pressure ∼3.91 MPa). In addition, the high-pressure CO_(2) adsorption characteristics of the southern Junggar Basin coal were characterized using a supercritical D-R adsorption model. Finally, the CO_(2) storage capacities in sub-bituminous coal under the in situ reservoir temperature and pressure were analyzed. Results indicated that the excess adsorption capacities increase gradually with increasing injection pressure before reaching an asymptotic maximum magnitude of ∼34.55 cm3/g. The supercritical D-R adsorption model is suitable for characterizing the excess/absolute CO_(2) adsorption capacity, as shown by the high correlation coefficients > 0.99. The CO_(2) adsorption capacity increases with declining temperature, indicating a negative effect of temperature on CO_(2) geological sequestration. By analyzing the statistical relationships of the D-R adsorption fitting parameters with the reservoir temperature, a CO_(2) adsorption capacity evolution model was established, which can be further used for predicting CO_(2) sequestration potential at in situ reservoir conditions. CO_(2) adsorption capacity slowly increases before reaching the critical CO_(2) density, following a rapid decrease at depths greater than ∼800 m in the southern Junngar Basin. The research results presented in this paper can provide guidance for evaluating CO_(2) storage potential in deep coal seams. | Sijian ZHENG Shuxun SANG Shiqi LIU Meng WANG Lutong CAO Xin JIN Guangjun FENG Yi YANG | 2023 | Frontiers of Earth Science2023,17,3: | 0
