| 1 | Proteomic Analysis of Macrophages:A Potential Way to Identify Novel Proteins Associated with Activation of Macrophages for Tumor Cell Killing显示文摘One major mechanism through which macrophages effectively kill tumor cells requires cell to cell contact,indicating that certain molecules expressed on cell surface of activated macrophages may mediate the tumoricidal capability. Tumor necrosis factor (TNF) and nitric oxide (NO) are the two classical mediators of tumor cell death. However,evidence of discrepancy is accumulating indicating these known mediators do not appear to account for the broad and potent tumoricidal activity of macrophages. To obtain a full repertoire of tumoricidal activation-associated membrane proteins,we combined one-dimensional SDS-PAGE with capillary liquid chromatography-tandem mass spectrometry (LC-MS/MS). Using this technique,we identified 454 activated macrophage specifically expressed proteins with extremely high confidence,including most known activation markers of macrophages,such as NO synthase (iNOS),Ym1,cyclooxygenase,etc. Membrane bound TNF-α was also identified on activated macrophages. However,it was also detected on thioglycolate elicited macrophages,indicating this molecule may not play a key role in conjugation-dependent tumor cell killing. In contrast,although NO has not been assigned as an effector molecule of conjugation-dependent tumoricidal pathway,iNOS was identified from membrane fraction of activated macrophages,suggesting NO may be involved in conjugation-dependent tumoricidal mechanism,because iNOS association with plasma membrane is ideally suited to deliver NO directly into the contacted tumor cells. | 3 | Inhibition of acetylcholinesterase by two genistein derivatives:kinetic analysis,molecular docking and molecular dynamics simulation显示文摘In this study two genistein derivatives(G1 and G2)are reported as inhibitors of acetylcholinesterase(AChE)and butyrylcholinesterase(BuChE),and differences in the inhibition of AChE are described.Although they differ in structure by a single methyl group,the inhibitory effect of G1(IC50¼264 nmol/L)on AChE was 80 times stronger than that of G2(IC50¼21,210 nmol/L).Enzyme-kinetic analysis,molecular docking and molecular dynamics(MD)simulations were conducted to better understand the molecular basis for this difference.The results obtained by kinetic analysis demonstrated that G1 can interact with both the catalytic active site and peripheral anionic site of AChE.The predicted binding free energies of two complexes calculated by the molecular mechanics/generalized born surface area(MM/GBSA)method were consistent with the experimental data.The analysis of the individual energy terms suggested that a difference between the net electrostatic contributions(ΔEele+ΔGGB)was responsible for the binding affinities of these two inhibitors.Additionally,analysis of the molecular mechanics and MM/GBSA free energy decomposition revealed that the difference between G1 and G2 originated from interactions with Tyr124,Glu292,Val294 and Phe338 of AChE.In conclusion,the results reveal significant differences at the molecular level in the mechanism of inhibition of AChE by these structurally related compounds. | Jiansong Fang Ping Wu Ranyao Yang Li Gao Chao Li Dongmei Wang Song Wu Ai-Lin Liu Guan-Hua Du | 2014 | Acta Pharmaceutica Sinica B2014,4,6: | 5 |

| 5 | Urban air quality, meteorology and traffic linkages: Evidence from a sixteen-day particulate matter pollution event in December 2015, Beijing显示文摘A heavy 16-day pollution episode occurred in Beijing from December 19, 2015 to January 3,2016. The mean daily AQI and PM_(2.5) were 240.44 and 203.6 μg/m^3. We analyzed the spatiotemporal characteristics of air pollutants, meteorology and road space speed during this period, then extended to reveal the combined effects of traffic restrictions and meteorology on urban air quality with observational data and a multivariate mutual information model. Results of spatiotemporal analysis showed that five pollution stages were identified with remarkable variation patterns based on evolution of PM_(2.5) concentration and weather conditions. Southern sites(DX, YDM and DS) experienced heavier pollution than northern ones(DL, CP and WL). | 7 | Extraordinary characteristics for one-dimensional parity-time-symmetric periodic ring optical waveguide networks显示文摘In this paper, we design a one-dimensional(1D) parity-time-symmetric periodic ring optical waveguide network(PTSPROWN) and investigate its extraordinary optical characteristics. It is found that quite different from traditional vacuum/dielectric optical waveguide networks, 1D PTSPROWN cannot produce a photonic ordinary propagation mode, but can generate simultaneously two kinds of photonic nonpropagation modes: attenuation propagation mode and gain propagation mode. It creates neither passband nor stopband and possesses no photonic band structure. | 20 | Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND experiment.
