Multiple genetic alterations and behavior of cellular biology in gastric cancer and other gastric mucosal lesions:H.pylori infection, histological types and staging显示文摘AIM To investigate the expression of multiplegenes and the behavior of cellular biology ingastric cancer(GC)and other gastric mucosallesions and their relations to Helicobacter pylori(H.pylori)infection, tumor staging andhistological subtypes.METHODS Three hundred and twenty-sevenspecimens of gastric mucosa obtained viaendoscopy or surgical resection, and ABCimmunohistochemical staining were used todetect the expression of p53,p16,Bcl-2 andCOX-2 proteins.H.pylori was determined byrapid urea test combined with pathologicalstaining or14C urea breath test.Cellular image analysis was performed in 66 patients withintestinal metaplasia(IM)and/or dysplasia(Dys). In 30 of them, both cancer and theparacancerous tissues were obtained at the timeof surgery.Histological pattern, tumor staging, lymph node metastasis, grading ofdifferentiation and other clinical data werestudied in the medical records.RESULTS p16 expression of IM or Dys wassignificantly lower in positive H.pylori chronicatrophic gastritis(CAG)than those withnegative H.pylori(CAG:54.8% vs 88.0%,IM:34.4% vs 69.6%,Dys:23.8% vs 53.6%,allP<0.05),Bcl-2 or COX-2 expression of IM orDys in positive H.pylori cases was significantlyhigher than that without H.pylori(Bcl-2:68.8%vs23.9%,90.5% vs 60.7%;COX-2:50.0% vs10.8%,61.8% vs 17.8%;all P<0.05).Themean number of most parameters of cellularimage analysis in positive H.pylori group wassignificantly higher than that in negative H.pylori group(Ellipser:53±14,40±12μm,Area1:748±572,302±202 μm2,Area2:3050±1661,1681±1990 μm2,all P<0.05;Ellipseb:79±23,58±15 μm,Ratio1:22%±5%,13%±4%,Ratio2:79%±17%,53%±20%,all P<0.01).There was significant correlation between Bcl-2and histologic pattern of gastric carcinoma, andbetween COX-2 and tumor staging or lymph nodemetastasis(Bcl-2:75.0% vs 16.7%;COX-2:76.0% vs 20.0%,79.2% vs 16.7%;allP<0.05).CONCLUSION p1l6, Bcl-2, and COX-2 but not p53 gene may play a role in the early genesis/ progression of gastric carcinoma and are associated with H. pylori infection. p53 gene is relatively late event in gastric tumorigenesis and mainly relates to its progression. There is more cellular-biological behavior of malignant tumor in gastric mucosal lesions with H. pylori infection. Aberrant Bcl-2 protein expression appears to be preferentially associated with the intestinal type cancer. COX-2 seems to be related to tumor staging and lymph node metastasis. | Heng Jun Gao Lian Zhen Yu Jian Feng Bai Yan Shen Peng Gu Sun Han Lin Zhao Kun Miu Xiu Zhen Lü Xiao Yong Zhang Zhi Quan Zhao | 2000 | World Journal of Gastroenterology2000,6,6: | 52

Pathological changes in the lungs and lymphatic organs of 12 COVID-19 autopsy cases显示文摘Systematic autopsy and comprehensive pathological analyses of COVID-19 decedents should provide insights into the disease characteristics and facilitate the development of novel therapeutics. In this study,we report the autopsy findings from the lungs and lymphatic organs of 12 COVID-19 decedents—findings that evaluated histopathological changes;immune cell signature and inflammatory factor expression in the lungs, spleen and lymph nodes. Here we show that the major pulmonary alterations included diffuse alveolar damage, interstitial fibrosis and exudative inflammation featured with extensive serous and fibrin exudates, macrophage infiltration and abundant production of inflammatory factors(IL-6,IP-10,TNFo?and IL-I f).The spleen and hilar lymph nodes contained lesions with tissue structure disruption and immune cell dysregulation, including lymphopenia and macrophage accumulation. These findings provide pathological evidence that links injuries of the lungs and lymphatic organs with the fatal systematic respiratory and immune malfunction in critically ill COVID-19 patients. | Qian Liu Yu Shi Jun Cai Yaqi Duan Rongshuai Wang Hongyan Zhang Qiurong Ruan Jiansha Li Lei Zhao Yifang Ping Rong Chen Liang Ren Xiaochun Fei Heng Zhang Rui Tang Xi Wang Tao Luo Xindong Liu Xuequan Huang Zhenhua Liu Qilin Ao Yong Ren Jing Xiong Zhicheng He Haibo Wu Wenjuan Fu Pengnan Zhao Xinwei Chen Guoqiang Qu Yunyun Wang Xi Wang Jia Liu Dongfang Xiang Sanpeng Xu Xiaowei Zhou Qingrui Li Jinghong Ma Heng Li Jie Zhang Sizhe Huang Xiaohong Yao Yiwu Zhou Chaofu Wang Dingyu Zhang Guoping Wang Liang Liu Xiu-Wu Bian | 2020 | National Science Review2020,7,12: | 6

2022 Chinese expert consensus and guidelines on clinical management of toxicity in anti-CD19 chimeric antigen receptor T-cell therapy for B-cell non-Hodgkin lymphoma显示文摘Adoptive cellular immunotherapy with chimeric antigen receptor(CAR)T cells has emerged as a novel modality for treating relapsed and/or refractory B-cell non-Hodgkin lymphoma(B-NHL).With increasing approval of CAR T-cell products and advances in CAR T cell therapy,CAR T cells are expected to be used in a growing number of cases. However,CAR T-cell-associated toxicities can be severe or even fatal, thus compromising the survival benefit from this therapy.Standardizing and studying the clinical management of these toxicities are imperative. In contrast to other hematological malignancies, such as acute lymphoblastic leukemia and multiple myeloma,anti-CD19 CAR T-cell-associated toxicities in B-NHL have several distinctive features, most notably local cytokine-release syndrome(CRS).However, previously published guidelines have provided few specific recommendations for the grading and management of toxicities associated with CAR T-cell treatment for B-NHL.Consequently,we developed this consensus for the prevention, recognition, and management of these toxicities,on the basis of published literature regarding the management of anti-CD19 CAR T-cell-associated toxicities and the clinical experience of multiple Chinese institutions. This consensus refines a grading system and classification of CRS in B-NHL and corresponding measures for CRS management, and delineates comprehensive principles and exploratory recommendations for managing anti-CD19 CAR T-cell-associated toxicities in addition to CRS. | Ping Li Yang Liu Yun Liang Jian Bo Sujun Gao Yongxian Hu Yu Hu He Huang Xiaojun Huang Hongmei Jing Xiaoyan Ke Jianyong Li Yuhua Li Qifa Liu Peihua Lu Heng Mei Ting Niu Yongping Song Yuqin Song Liping Su Sanfang Tu Jianxiang Wang Depei Wu Zhao Wang Kailin Xu Zhitao Ying Qingming Yang Yajing Zhang Fengxia Shi Bin Zhang Huilai Zhang Xi Zhang Mingfeng Zhao Weili Zhao Xiangyu Zhao Liang Huang Jun Zhu Wenbin Qian Weidong Han Aibin Liang | 2023 | Cancer Biology & Medicine2023,20,2: | 3

Panda X-III: Searching for neutrinoless double beta decay with high pressure ^(136)Xe gas time projection chambers显示文摘Searching for the neutrinoless double beta decay(NLDBD)is now regarded as the topmost promising technique to explore the nature of neutrinos after the discovery of neutrino masses in oscillation experiments.Panda X-III(particle and astrophysical xenon experiment III)will search for the NLDBD of136Xe at the China Jin Ping Underground Laboratory(CJPL).In the first phase of the experiment,a high pressure gas Time Projection Chamber(TPC)will contain 200 kg,90%136Xe enriched gas operated at10 bar.Fine pitch micro-pattern gas detector(Microbulk Micromegas)will be used at both ends of the TPC for the charge readout with a cathode in the middle.Charge signals can be used to reconstruct the electron tracks of the NLDBD events and provide good energy and spatial resolution. The detector will be immersed in a large water tank to ensure~5 m of water shielding in all directions. The second phase,a ton-scale experiment, will consist of five TPCs in the same water tank, with improved energy resolution and better control over backgrounds. | Xun Chen Chang Bo Fu Javier Galan Karl Giboni Franco Giuliani Ling Hui Gu Ke Han Xiang Dong Ji Heng Lin Jiang Lai Liu Kai Xiang Ni Hiroki Kusano Xiang Xiang Ren Shao Bo Wang Yong Yang Dan Zhang Tao Zhang Li Zhao Xiang Ming Sun Shou Yang Hu Si Yu Jian Xing Long Li Xiao Mei Li Hao Liang Huan Qiao Zhang Ming Rui Zhao Jing Zhou Ya Jun Mao Hao Qiao Si Guang Wang Ying Yuan Meng Wang Amir N.Khan Neill Raper Jian Tang Wei Wang Jia Ning Dong Chang Qing Feng Cheng Li Jian Bei Liu Shu Bin Liu Xiao Lian Wang Dan Yang Zhu Juan F.Castel Susana Cebri'an Theopisti Dafni Javier G.Garza Igor G.Irastorza Francisco J.Iguaz Gloria Luz ′on Hector Mirallas Stephan Aune Eric Berthoumieux Yann Bedfer Denis Calvet Nicole d'Hose Alain Delbart Maria Diakaki Esther Ferrer-Ribas Andrea Ferrero Fabienne Kunne Damien Neyret Thomas Papaevangelou Franck Sabatie Maxence Vanderbroucke AnDi Tan Wick Haxton Yuan Mei Chinorat Kobdaj Yu-Peng Yan | 2017 | Science China(Physics,Mechanics & Astronomy)2017,60,6: | 2
Uncertainty analysis and robust design optimization for the heat-assisted bending of high-strength titanium tube显示文摘Heat-assisted rotary draw bending(HRDB)is a promising technique for manufacturing difficult-to-form tubular components comprising high-strength titanium tubes(HSTTs)with small bending radii. However,as a multidie constrained and thermomechanical coupled process with many uncertainty factors,a high risk of several defects, such as cross-section distortion, over wall thinning,or even cracking,is present.Achieving the robust design optimization(RDO)of complex forming processes remains a nontrivial and challenging scientific issue.Herein, considering a high-strength Ti-3Al-2.5V titanium alloy tube as a case material, the five significant uncertainty factors in HRDB,i.e.,temperature distribution, tube geometrical characteristics, tube material parameters,tube/tool friction, and boost velocity had been analyzed.Subsequently, considering the preheating and HRDB of HSTT,a whole-process thermomechanical three-dimensional finite element model was established and validated for virtual experiments.Further, considering the maximum section distortion Q and maximum wall-thickness thinning t as the optimization objectives and the mean and variance of material and forming parameters,an RDO model was established. Finally, the Pareto optimal solutions were obtained using the nondominated sorting genetic algorithm II,and a minimum distance selection method was employed to obtain the satisfactory solution. Results show that the optimized solutions considering the uncertainty factors reduce the maximum section distortion rate of HSTT after bending by 38.1%and the maximum wallthickness thinning rate by 27.8%. | ZHANG Zhao YANG JingChao HUANG WeiLiang MA Jun LI Heng | 2021 | Science China(Technological Sciences)2021,64,10: | 1

Metabolic uncoupler,3,3’,4’,5-tetrachlorosalicylanilide addition for sludge reduction and fouling control in a gravity-driven membrane bioreactor显示文摘The gravity-driven membrane bioreactor(MBR)system is promising for decentralized sewage treatment because of its low energy consumption and maintenance requirements. However, the growing sludge not only increases membrane fouling, but also augments operational complexities(sludge discharge). We added the metabolic uncoupler 3,3’,4f,5-tetrachlorosalicylanilide(TC$)to the system to deal with the mentioned issues.Based on the results,TCS addition effectively decreased sludge ATP and sludge yield(reduced by 50%).Extracellular polymeric substances(EPS;proteins and polysaccharides)decreased with the addition of TCS and were transformed into dissolved soluble microbial products(SMPs)in the bulk solution, leading to the break of sludge floes into small fragments.Permeability was increased by more than two times, reaching 60-70 L/m2/h bar when 10-30 mg/L TCS were added, because of the reduced suspended sludge and the formation of a thin cake layer with low EPS levels.Resistance analyses confirmed that appropriate dosages of TCS primarily decreased the cake layer and hydraulically reversible resistances.Permeability decreased at high dosage(50 mg/L)due to the release of excess sludge fragments and SMP into the supernatant, with a thin but more compact fouling layer with low bioactivity developing on the membrane surface, causing higher cake layer and pore blocking resistances. Our study provides a fundamental understanding of how a metabolic uncoupler affects the sludge and bio-fouling layers at different dosages, with practical relevance for in situ sludge reduction and membrane fouling alleviation in MBR systems. | An Ding Yingxue Zhao Huu Hao Ngo Langming Bai Guibai Li Heng Liang Nanqi Ren Jun Nan | 2020 | Frontiers of Environmental Science & Engineering2020,14,6: | 1
Endocrine Disruption of Cadmium in Rats Using the OECD Enhanced TG 407 Test System显示文摘Objective To evaluate the endocrine disrupting effects of cadmium(Cd) using OECD enhanced TG407 test guideline. Methods Sprague-Dawley(SD) rats were randomly divided into six groups and accordingly administered with 0, 1, 2.5, 5, 10, 20 mg/kg·BW/day of Cd by gavage for 28 days. Body weight, food consumption, hematology, biochemistry, sex hormone levels, urinary β2-microglobulin, organ weights and histopathology and estrous cycle were detected. Results Cd could significantly decrease animals' body weight(P<0.05). Serum luteinizing hormone(LH) at 10-20 mg/kg·BW groups and testosterone(T) at 2.5 and 10 mg/kg·BW groups decreased significantly(P<0.05). However, no statistically significant change was found in urinary β2-microglobulin among Cd-treatment groups(P>0.05). Endpoints related to female reproduction including uterus weight and histopathological change at 10-20 mg/kg·BW groups showed significant increase(P<0.05). While among male rats in 2.5, 10, 20 mg/kg·BW groups, weight of prostate, thyroids, and seminal vesicle glands significantly decreased(P<0.05). Moreover, no histopathological change was observed in kidney. Conclusion Results suggested that Cd can cause endocrine disrupting effects in SD rats. Comparing with possible renal toxicity of Cd, its toxicity on endocrine system was more sensitive. | WANG Heng Juan LIU Zhao Ping JIA Xu Dong CHEN Hao TAN Yan Jun | 2014 | Biomedical and Environmental Sciences2014,27,12: | 1
Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector. Some of JUNO's features make it an excellent location for^8B solar neutrino measurements, such as its low-energy threshold, high energy resolution compared with water Cherenkov detectors, and much larger target mass compared with previous liquid scintillator detectors. In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable, assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition, approximately 60,000 signal and 30,000 background events are expected. This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter, which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle. Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better, depending on the central value, and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND experiment. | 18 | Template-based knowledge reuse method for generating high-speed railway virtual construction scenes显示文摘Virtual construction has become an important approach to the high-quality development of high-speed railways, but existing methods have problems such as low efficiency in generating virtual construction scenes and the inability to reuse construction knowledge. To support the rapid visual representation of multiple types of construction processes and construction methods,a template-based knowledge reuse method is proposed. The method includes using a component-based modeling mode to build body structure models of a high-speed railway project and generate a 3D scene;decomposing the construction process and building a construction process knowledge base;establishing joint linkage models of construction machinery and forming a construction method knowledge template;and fusing multiple types of information according to a time sequence to visualize the construction process.Based on the template-based knowledge reuse method,a prototype system was developed, and virtual construction experiments were carried out.The results show that this method achieves the reuse of construction knowledge at different levels including construction machinery level, construction method level, and work site level.Compared with animation software for virtual construction, this method improves the production efficiency by 87%.Moreover,this method can provide a multilevel knowledge reuse scheme for diversified virtual construction. | Heng Zhang Wen Zhao Zujie Han Jun Zhu Qing Zhu Xinwen Ning Dengke Fan Hua Wang Fengpin Jia Wei Fang Bin Yang Weilian Li | 2023 | International Journal of Digital Earth2023,16,1: | 0
Conformational effect on fluorescence emission of tetraphenylethylene-based metallacycles显示文摘Herein,we designed and constructed two metallacycles,1 and 2,to illustrate the conformational effect of isomeric AIE fluoropho res on the platfo rm of supramolecular coordination complexe s(SCCs).Specifically, the dangling phenyl rings in TPE units of the metallacycle 1 align completely outside the main cyclic structure, while in the metallacycle 2,these phenyl rings align half inside and half outside. The experimental results showed that two metallacycles exhibited different behaviors in terms of AIE fluorescence and chemical sensing, which could be attributed to the subtle structural difference of the TPE units. This work repre sents the unification of topics such as self-assembly,AIE,and chemical sensing, and further promotes the understanding for the structure-property relationship of isomeric AIE fluorophores. | Zhewen Guo Jun Zhao Yuhang Liu Guangfeng Li Heng Wang Yali Hou Mingming Zhang Xiaopeng Li Xuzhou Yan | 2021 | Chinese Chemical Letters2021,32,5: | 0
