1Chromobox homolog 4 overexpression inhibits TNF-α-induced matrix catabolism and senescence by suppressing activation of NF-κB signaling pathway in nucleus pulposus cells显示文摘Intervertebral disc degeneration(IDD)is featured as enhanced catabolism of extracellular matrix(ECM)in the nucleus pulposus(NP),in which tumor necrosis factor-alpha(TNF-α)-related cell senescence is involved.Chromobox homolog protein 4(CBX4)exhibits anti-inflammatory effects and shows promising therapeutic potential.Thus,in the present study,we explore the role of CBX4 in IDD.Immunohistochemistry staining reveals that CBX4 expression is decreased in severe degenerative NP tissues compared to mild degenerative tissues,and real-time PCR and western blot analysis results show that CBX4 expression is downregulated under TNF-αstimulation in NP cells.siRNA and adenoviruses are used to knockdown or overexpress CBX4,respectively.The results demonstrate that CBX4 knockdown augments the catabolism of ECM in human NP cells,while CBX4 overexpression in rat NP cells restores the ECM degradation induced by TNF-α,as illustrated by immunofluorescence and western blot analysis.In addition,transcriptome sequencing results reveal the regulatory effect of CBX4 on the cell cycle,and further western blot analysis and senescence-associatedβ-galactosidase staining assay indicate that CBX4 overexpression alleviates cell senescence in the presence of TNF-α.Moreover,the phosphorylation of p65,which indicates the activation of NF-κB signaling,is measured by western blot analysis and immunofluorescence assay,and the results reveal that CBX4 overexpression reduces the TNF-α-induced increase in the p-p65/p65 ratio.In addition,the effect of CBX4 overexpression in NP cells is suppressed by NF-κB agonist.In summary,our results indicate that CBX4 overexpression can suppress TNF-α-induced matrix catabolism and cell senescence in the NP by inhibiting NF-κB activation.This study may provide new approaches for preventing and treating IDD.Yangyang Zhang Shuangxing Li Junmin Hong Jiansen Yan Zhengqi Huang Jiajun Wu Zhihuai Deng Tianyu Qin Kang Xu Wei Ye 2022Acta Biochimica et Biophysica Sinica2022,54,7:2

4Effects of Zhichan powder on signal transduction and apoptosis-associated gene expression in the substantia nigra of Parkinson's disease rats显示文摘Previous studies have shown that Zhichan powder elevated immunity and suppressed oxidation in mice. Rat models of Parkinson's disease were induced by stereotaxically injecting 6-hydroxydopamine into the substantia nigra. The rat models were intragastrically treated with Zhichan powder, which is composed of milkvetch root, ginseng, bunge swallowwort root, himalayan teasel root, Magnolia officinalis, Ligustrum lucidum Ait. and szechwan lovage rhizome. Immunohistochemistry and reverse transcription-PCR results demonstrated that mRNA and protein expression of tumor necrosis factor receptor 1, Fas, caspase-8, cytochrome C, Bax, caspase-3, and p53 significantly increased, but Bcl-2 expression significantly decreased in the substantia nigra of rats with Parkinson's disease. Following Zhichan powder administration, mRNA and protein expression of tumor necrosis factor receptor 1, Fas, caspase-8, cytochrome C, Bax, caspase-3, and p53 diminished, but Bcl-2 expression increased in the rat substantia nigra. 6Structural phase transition and dielectric switching in an organic-inorganic hybrid rare-earth double perovskite-type compound:(DMP)_(2)LaRb(NO_(3))_(6)(DMP=N,N-dimethylpyrrolidinium cation)显示文摘In recent years,it has been found that the flexibility of structure and diversity of the components endow quite an amount of the organic-inorganic hybrid perovskites with novel properties,i.e.,structural phase transitions.Considering the natural advantage of the perovskite-type structure in generation of stimuliresponsive or smart materials,we synthesized an organic-inorganic hybrid rare-earth double perovskitetype compound,(DMP)_(2) LaRb(NO_(3))_(6)(DMP=N,N-dimethylpyrrolidinium cation,1).It shows reversible phase transition at 219/209 K(heating/cooling).Variable-temperature single-crystal structure analysis and dielectric constant measurements reveal that the thermal vibrations of the polar cation guests and the distortion of the anionic cage-like framework are the origin of the phase transition.Meanwhile,the movement of polar cation in crystal lattices arouses dielectric transition between the low-and highdielectric states,resulting in a switchable property of dielectric constant.The results reveal that the rare-earth double perovskite provides a promising platform for achieving switchable physical/chemical properties.Jiajun Ma Qi Xu Le Ye Qinwen Wang Zhixin Gong Chao Shi Hengyun Ye Yi Zhang 2022Journal of Rare Earths2022,40,6:0
7Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND 9Sodium butyrate activates HMGCS2 to promote ketone body production through SIRT5-mediated desuccinylation显示文摘Ketone bodies have beneficial metabolic activities,and the induction of plasma ketone bodies is a health promotion strategy.Dietary supplementation of sodium butyrate(SB)is an effective approach in the induction of plasma ketone bodies.However,the cellular and molecular mechanisms are unknown.In this study,SB was found to enhance the catalytic activity of 3-hydroxy-3-methylglutaryl-CoA synthase 2(HMGCS2),a rate-limiting enzyme in ketogenesis,to promote ketone body production in hepatocytes.SB administrated by gavage or intraperitoneal injection significantly induced bloodβ-hydroxybutyrate(BHB)in mice.BHB production was induced in the primary hepatocytes by SB.Protein succinylation was altered by SB in the liver tissues with down-regulation in 58 proteins and up-regulation in 26 proteins in the proteomics analysis.However,the alteration was mostly observed in mitochondrial proteins with 41%down-and 65%up-regulation,respectively.Succinylation status of HMGCS2 protein was altered by a reduction at two sites(K221 and K358)without a change in the protein level.The SB effect was significantly reduced by a SIRT5 inhibitor and in Sirt5-KO mice.The data suggests that SB activated HMGCS2 through SIRT5-mediated desuccinylation for ketone body production by the liver.The effect was not associated with an elevation in NAD+/NADH ratio according to our metabolomics analysis.The data provide a novel molecular mechanism for SB activity in the induction of ketone body production.Yanhong Xu Xiaotong Ye Yang Zhou Xinyu Cao Shiqiao Peng Yue Peng Xiaoying Zhang Yili Sun Haowen Jiang Wenying Huang Hongkai Lian Jiajun Yang Jia Li Jianping Ye 2023Frontiers of Medicine2023,17,2:0
10The Construction of an Intrauterine Diagnosis and Treatment System and Comprehensive Lifecycle Health Service of Congenital Heart Disease:Xinhua Hospital Model显示文摘With the growing influence of slow population growth and population aging,China has established the birth policy and issued a series of documents to promote maternal and fetal health and improve the birth rate.With the increase in prevalence of birth defects,timely diagnosis and intervention in utero provide possibilities to reduce unnecessary abortions and offer better prognosis.Congenital heart disease(CHD),as one of the most common congenital birth defects,is the leading cause of mortality in patients aged<5 years,and brings a heavy burden to both the affected families and society.Fetuses with CHD are associated with an increased risk of pregnancy-related complications and premature birth,and children with CHD typically face growth and developmental problems even after the correction of malformation.Therefore,management including diagnosis,treatment,and rehabilitation throughout the fetal period into childhood and even adulthood is essential for children with CHD.Based on the rapid advances in intrauterine and perinatal medicine and an in-depth collaboration among obstetrics and pediatrics,a novel diagnosis and treatment system has been established for the management of CHD in the past 2 decades in Shanghai Xinhua Hospital.This Intrauterine Diagnosis and Treatment System and Comprehensive Lifecycle Health Service of Congenital Heart Disease model provides prenatal diagnosis,intrauterine intervention,delivery room service and neonatal therapies,and postintrauterine rehabilitation for children with CHD.We have developed a four-dimensional spatiotemporal image correlation echocardiography and a three-dimensional cardiac virtual endoscopy system for the intrauterine diagnosis of CHD,dramatically raising the diagnostic utility.Our innovative and independent newborn-intervention technique has effectively reduced the re-intervention rate in patients with pulmonary atresia with intact ventricular septum and critical pulmonary stenosis.In 2018,Xinhua Hospital independently performed the case of fetal aortic valvuloplasty in Asia through a multidepartment collaborative effort.All children treated in this system achieved biventricular circulation and a better long-term postoperative outcome.We also have conducted postoperative rehabilitation therapy to promote the development and health of children with CHD.The practice of Xinhua model has reduced unnecessary abortion of CHD fetuses,reduced the mortality rate associated with critical CHD,and improved the mid-and long-term prognosis in CHD,which is essential to promote the fertility level and children’s health.Furthermore,translational medicine platform and the birth cohort Early Life Plan was constructed to explore the origins of major developmental diseases and establish an early intervention model in CHD.This practice of assessment of the intrauterine system has been expanded to other congenital defects in Xinhua Hospital,and sequential treatment of more than 2,000 cases has been completed to date.Based on practice in intrauterine management of CHD and other diseases,the concept of Intrauterine Pediatrics was proposed as a first to emphasize early prevention and intervention of childhood diseases and promote a comprehensive lifecycle service for children.The development and evolution of this system requires further attention not only from researchers but also from the government and global medical communities.Shiwei Jiang Jiajun Ye Hualin Wang Jian Wang Sun Cheni Yongjun Zhang Qing Du Ling Yang Lei Wang Kun Sun 2023Cardiology Discovery2023,3,3:0
11Magma Mixing as a Trigger for Sulphide Saturation in the UG2 Chromitite(Bushveld):Evidence from the Silicate and Sulphide Melt Inclusions in Chromite显示文摘It is of great importance to understand the origin of UG2 chromitite reefs and reasons why some chromitite reefs contain relatively high contents of platinum group elements(PGEs: Os, Ir, Ru, Rh,Pt, Pd) or highly siderophile elements(HSEs: Au, Re, PGE). This paper documents sulphide-silicate assemblages enclosed in chromite grains from the UG2 chromitite. These are formed as a result of crystallisation of sulphide and silicate melts that are trapped during chromite crystallisation. The inclusions display negative crystal shapes ranging from several micrometres to 100 μm in size.Interstitial sulphide assemblages lack pyrrhotite and consist of chalcopyrite, pentlandite and some pyrite. The electron microprobe data of these sulphides show that the pentlandite grains present in some of the sulphide inclusions have a significantly higher iron(Fe) and lower nickel(Ni) content than the pentlandite in the rock matrix. Pyrite and chalcopyrite show no difference. The contrast in composition between inter-cumulus plagioclase(An_(68)) and plagioclase enclosed in chromite(An_(13)), as well as the presence of quartz, is consistent with the existence of a felsic melt at the time of chromite saturation.Detailed studies of HSE distribution in the sulphides and chromite were conducted by LA-ICP-MS(laser ablation-inductively coupled plasma-mass spectrometry), which showed the following.(Ⅰ) Chromite contained no detectable HSE in solid solution.(Ⅱ) HSE distribution in sulphide assemblages interstitial to chromite was variable. In general, Pd, Rh, Ru and Ir occurred dominantly in pentlandite, whereas Os,Pt and Au were detected only in matrix sulphide grains and were clearly associated with Bi and Te.(Ⅲ)In the sulphide inclusions,(a) pyrrhotite did not contain any significant amount of HSE,(b) chalcopyrite contained only some Rh compared to the other sulphides,(c) pentlandite was the main host for Pd,(d)pyrite contained most of the Ru, Os, Ir and Re,(e) Pt and Rh were closely associated with Bi forming a continuous rim between pyrite and pentlandite and(f) no Au was detected. 12NF-κB regulates brown adipocyte function through suppression of ANT2显示文摘The transcription factor nuclear factor of kappa-light-chain-enhancer of activated B cells(NF-κB)is expressed in brown adipocytes,but its role remains largely unknown in the cells.This issue was addressed in current study by examining NF-κB in brown adipocytes in vitro and in vivo.NF-κB activity was increased by differentiation of brown adipocytes through elevation of p65(RelA)expression.The transcriptional activity of NF-κB was induced by the cold stimulation with an elevation in S276 phosphorylation of p65 protein.Inactivation of NF-κB in brown adipocytes made the knockout mice[uncoupling protein 1(Ucp1)-CreER-p65^(f/f),U-p65-KO]intolerant to the cold environment.The brown adipocytes exhibited an increase in apoptosis,a decrease in cristae density and uncoupling activity in the interscapular brown adipose tissue(iBAT)of p65-KO mice.The alterations became severer after cold exposure of the KO mice.The brown adipocytes of mice with NF-κB activation(p65 overexpression,p65-OE)exhibited a set of opposite alterations with a reduction in apoptosis,an increase in cristae density and uncoupling activity.In mechanism,NF-κB inhibited expression of the adenine nucleotide translocase 2(ANT2)in the control of apoptosis.Data suggest that NF-κB activity is increased in brown adipocytes by differentiation and cold stimulation to protect the cells from apoptosis through down-regulation of ANT2 expression.Shiqiao Peng Xiaoying Zhang Lili Yu Yanhong Xu Yang Zhou Shengnan Qian Xinyu Cao Xiaotong Ye Jiajun Yang Weiping Jia Jianping Ye 2022Acta Pharmaceutica Sinica B2022,12,3:0
13Asymmetric side-chain engineering of organic semiconductor for ultrasensitive gas sensing显示文摘Molecular structure of organic semiconductor plays a critical role in determining the performance and functionality of organic electronic devices,by optimizing the electrical,optical and physicochemical properties.Substituted alkyl chains are fundamental units in tailering the solubility and assemblability,among which the asymmetric properties have been reported as key element for controlling the packing motifs and intrinsic charge transport.Here,we expanded the scope of molecular asymmetry dependent sensing features based on a new series of naphthalene diimides(NDI)-based derivatives substituted with a same branching alkyl chain but various linear-shaped alkyl chains(Cn-).A clear molecular stacking change,from head-to-head bilayer to head-to-tail monolayer packing model,is observed based on the features of anisotropic molecular interactions with the change in the chain length.Most importantly,a unique LUMO level shift of 0.17 eV is validated for NDI-PhC4,providing a record sensitivity up to 150%to 0.01 ppb ammonia,due to the desired molecular reactivity and device amplification properties.These results indicate that asymmetric side-chain engineering opens a route for breath healthcare.Xiaoying Ma Xiaojuan Dai Lanyi Xiang Jiajun Chang Danfeng Zhi Haozhen Zhao Zhenjie Ni Ye Zou Xike Gao Fengjiao Zhang 2024Chinese Chemical Letters2024,35,3:0
14Integrated proteogenomic characterization across major histological types of pituitary neuroendocrine tumors显示文摘Pituitary neuroendocrine tumor(PitNET)is one of the most common intracranial tumors.Due to its extensive tumor heterogeneity and the lack of high-quality tissues for biomarker discovery,the causative molecular mechanisms are far from being fully defined.Therefore,more studies are needed to improve the current clinicopathological classification system,and advanced treatment strategies such as targeted therapy and immunotherapy are yet to be explored.Here,we performed the largest integrative genomics,transcriptomics,proteomics,and phosphoproteomics analysis reported to date for a cohort of 200 PitNET patients.Genomics data indicate that GNAS copy number gain can serve as a reliable diagnostic marker for hyperproliferation of the PIT1 lineage.Proteomics-based classification of PitNETs identified 7 clusters,among which,tumors overexpressing epithelial-mesenchymal transition(EMT)markers clustered into a more invasive subgroup.Further analysis identified potential therapeutic targets,including CDK6,TWIST1,EGFR,and VEGFR2,for different clusters.Immune subtyping to explore the potential for application of immunotherapy in PitNET identified an association between alterations in the JAK1-STAT1-PDL1 axis and immune exhaustion,and between changes in the JAK3-STAT6-FOS/JUN axis and immune infiltration.These identified molecular markers and alternations in various clusters/subtypes were further confirmed in an independent cohort of 750 PitNET patients.This proteogenomic analysis across traditional histological boundaries improves our current understanding of PitNET pathophysiology and suggests novel therapeutic targets and strategies.Fan Zhang Qilin Zhang Jiajun Zhu Boyuan Yao Chi Ma Nidan Qiao Shiman He Zhao Ye Yunzhi Wang Rui Han Jinwen Feng Yongfei Wang Zhaoyu Qin Zengyi Ma Kai Li Yichao Zhang Sha Tian Zhengyuan Chen Subei Tan Yue Wu Peng Ran Ye Wang Chen Ding Yao Zhao 2022Cell Research2022,32,12:0
15Rosuvastatin suppresses TNF-α-induced matrix catabolism,pyroptosis and senescence via the HMGB1/NF-κB signaling pathway in nucleus pulposus cells显示文摘Intervertebral disc degeneration is mainly caused by irregular matrix metabolism in nucleus pulposus cells and involves inflammatory factors such as TNF-α.Rosuvastatin,which is widely used in the clinic to reduce cholesterol levels,exerts anti-inflammatory effects,but whether rosuvastatin participates in IDD remains unclear.The current study aims to investigate the regulatory effect of rosuvastatin on IDD and the potential mechanism.In vitro experiments demonstrate that rosuvastatin promotes matrix anabolism and suppresses catabolism in response to TNF-αstimulation.In addition,rosuvastatin inhibits cell pyroptosis and senescence induced by TNF-α.These results demonstrate the therapeutic effect of rosuvastatin on IDD.We further find that HMGB1,a gene closely related to cholesterol metabolism and the inflammatory response,is upregulated in response to TNF-αstimulation.HMGB1 inhibition or knockdown successfully alleviates TNF-α-induced ECM degradation,senescence and pyroptosis.Subsequently,we find that HMGB1 is regulated by rosuvastatin and that its overexpression abrogates the protective effect of rosuvastatin.We then verify that the NF-κB pathway is the underlying pathway regulated by rosuvastatin and HMGB1.In vivo experiments also reveal that rosuvastatin inhibits IDD progression by alleviating pyroptosis and senescence and downregulating HMGB1 and p65.This study might provide new insight into therapeutic strategies for IDD.Weijian Chen Zhihuai Deng Jianxiong Zhu Liang Yuan Shuangxing Li Yangyang Zhang Jiajun Wu Zhengqi Huang Tianyu Qin Wei Ye 2023Acta Biochimica et Biophysica Sinica2023,55,5:0
