| 4 | Influence of continuous veno-venous hemofiltration on the course of acute pancreatitis显示文摘AIM: To investigate whether continuous veno-venous hemofiltration (CWH) in different filtration rate to eliminate cytokines would result in different efficiency in acute pancreatitis, whether the saturation time of filter membrane was related to different filtration rate, and whether the onset time of CWH could influence the survival of acute pancreatitis.METHODS: Thirty-seven patients were classified into four groups randomly. Group 1 underwent low-volume CVVH within 48 h of the onset of abdominal pain (early CVVH,n = 9). Group 2 received low-volume CVVH after 96 h of the onset of abdominal pain (late CVVH, n = 10). Group 3underwent high-volume CWH within 48 h of the onset of abdominal pain (early CWH, n = 9). Group 4 received high-volume CWH after 96 h of theonset of abdominal pain (late CVVH, n = 9). CVVH was sustained for at least 72 h. Blood was taken before hemofiltration, and ultrafiltrate was collected at the start of CWH and every 12 h during CVVH period for the purpose of measuring the concentrations of TNF-α, IL-1β and IL-6. The concentrations of TNF-α, IL-1β and IL-6 were measured by swine-specific El ISA. The Solartron 1 255 B frequency response analyzer (British) was used to observe the resistance of filter membrane.RESULTS: The survival rate had a significant difference (94.44% vs68.42%, P<0.01) high-volume and low-volume CVVH patients. The survival rate had also a significant difference (88.89% vs 73.68%, P<0.05) between early and late CWH patients. The hemodynamic deterioration (MAP, HR, CVP) was less severe in groups 4 and 1 than that in group 2, and in group 3 than in group 4. The adsorptive saturation time of filters membranes was 120-180 min if the filtration rate was 1 000-4 000 mL/h. After the first, second and third new hemofilters were changed,serum TNF-α concentrations had a negative correlation with resistance (r: -0.91, -0.89, and -0.86, respectively in group 1; -0.89, -0.85, and -0.76, respectively in group 2;-0.88, -0.92, and -0.82, respectively in group 3; -0.84,-0.87, and -0.79, respectively in group 4). The decreasing extent of TNF-α, IL-1β and IL-6 was significantly different between group 3 and group 1 (TNF-α P<0.05, IL-1β P<0.05,IL-6 P<0.01), between group 4 and group 2 (TNF-α P<0.05,IL-1β P<0.05, IL-6 P<0.01), between group 1 and group 2(TNF-α P<0.05, IL-1β P<0.05, IL-6 P<0.05), and between group 3 and group 4 (TNF-α P<0.01, IL-1β P<0.01, IL-6P<0.05),respectively during CVVH period. The decreasing extent of TNF-α and IL-1β was also significantly different between survival patients and dead patients (TNF-αP<0.05, IL-1β P<0.05).In survival patients, serum concentration of TNF-α and IL-1β decreased more significantly than that in dead patients.CONCLUSION: High-volume and early CWH improve hemodynamic deterioration and survival in acute pancreatitis patients. High-volume CVVH can eliminate cytokines more efficiently than low-volume CWH. The survival rate is related to the decrease extent of TNF-α and IL-1β.The adsorptive saturation time of filter membranes are different under different filtration rate condition. | 6 | Effects of dietary protein/energy ratio on growth performance,carcass trait,meat quality,and plasma metabolites in pigs of different genotypes显示文摘Background:The protein/energy ratio is important for the production performance and utilization of available feed resources by animals.Increased protein consumption by mammals leads to elevated feed costs and increased nitrogen release into the environment.This study aimed to evaluate the effects of dietary protein/energy ratio on the growth performance,carcass traits,meat quality,and plasma metabolites of pigs of different genotypes.Methods:Bama mini-pigs and Landrace pigs were randomly assigned to two dietary treatment groups(Chinese conventional diet with low protein/energy ratio or National Research Council diet with high protein/energy ratio;n = 24 per treatment) in a 2x2 factorial arrangement.Blood and muscle samples were collected at the end of the nursery,growing,and finishing phases.Results:We observed significant interactions(P< 0.05) between breed and diet for total fat percentage,intramuscular fat(IMF) content,protein content in biceps femoris(BF) muscle,and plasma urea nitrogen(UN) concentration in the nursery phase;for average daily gain(ADG),average daily feed intake(ADFI),dry matter,IMF content in psoas major(PM) muscle,and plasma total protein and albumin concentrations in the growing phase;and for drip loss and plasma UN concentration in the finishing phase.Breed influenced(P < 0.05) growth performance,carcass traits,and meat quality,but not plasma metabolites.Throughout the trial,Landrace pigs showed significantly higher(P < 0.05) ADG,ADFI,dressing percentage,lean mass rate,and loin-eye area than did Bama mini-pigs,but significantly lower(P< 0.05)feed/gain ratio,fat percentage,backfat thickness,and IMF content.Dietary protein/energy ratio influenced the pH value,chemical composition of BF and PM muscles,and plasma activities of glutamic-pyruvic transaminase and gamma-glutamyl transpeptidase,and plasma concentration of UN.Conclusions:Compared with Landrace pigs,Bama mini-pigs showed slower growth and lower carcass performance,but had better meat quality.Moreover,unlike Landrace pigs,the dietary protein/energy ratio did not affect the growth performance of Bama mini-pigs.These results suggest that,in swine production,low dietary protein/energy ratio may be useful for reducing feed costs and minimizing the adverse effects of ammonia release into the environment. | Yingying Liu Xiangfeng Kong Guoli Jiang Bi'e Tan Jinping Deng Xiaojian Yang Fengna Li Xia Xiong Yulong Yin | 2015 | Journal of Animal Science and Biotechnology2015,6,4: | 37 |
| 7 | Ultrafast epitaxial growth of metre-sized single-crystal graphene on industrial Cu foil显示文摘A foundation of the modern technology that uses single-crystal silicon has been the growth of highquality single-crystal Si ingots with diameters up to 12 inches or larger. For many applications of graphene, large-area high-quality(ideally of single-crystal) material will be enabling. Since the first growth on copper foil a decade ago, inch-sized single-crystal graphene has been achieved. We present here the growth, in 20 min, of a graphene film of(5 ×50) cm^2 dimension with >99% ultra-highly oriented grains.This growth was achieved by:(1) synthesis of metre-sized single-crystal Cu(1 1 1) foil as substrate;(2)epitaxial growth of graphene islands on the Cu(1 1 1) surface;(3) seamless merging of such graphene islands into a graphene film with high single crystallinity and(4) the ultrafast growth of graphene film.These achievements were realized by a temperature-gradient-driven annealing technique to produce single-crystal Cu(1 1 1) from industrial polycrystalline Cu foil and the marvellous effects of a continuous oxygen supply from an adjacent oxide. The as-synthesized graphene film, with very few misoriented grains(if any), has a mobility up to ~23,000 cm^2 V^(-1)s^(-1)at 4 K and room temperature sheet resistance of ~230 Ω/□. | 8 | 42,573 cases of hepatectomy in China: a multicenter retrospective investigation显示文摘Hepatectomy is currently routinely performed in most hospitals in China. China owns the largest population of liver diseases and the biggest number of liver resection cases. A nationwide multicenter retrospective investigation involving 112 hospitals was performed, and focused on liver resection for patients with hepatocellular carcinoma(HCC). 42,573 cases of hepatectomy were enrolled, and 18,275 valid cases of liver resection for HCC patients were selected for statistical analysis. The epidemiology of HCC, distribution of hepatectomy, postoperative complications and prognosis were finally analyzed. In the 18,275 HCC patients,81% had hepatitis B virus infection and 10% had hepatitis C virus infection. 38% of the HCC patients had normal Alphafetoprotein(AFP) level, and other 35% had an AFP level lower than 400 ng mL^(-1). In the study period, 97% of the hepatectomy for HCC were treated with open surgery, and 23.81% had vascular exclusion techniques. The operation time was(191.7±105.6) min,the blood loss was(546.0±562.8) m L, and blood transfusion was(543.0±1,035.2) m L. The median survival for HCC patients was 631 days, with 1-, 3-, and 5-year overall survival of 73.2%, 28.8% and 19.6%, respectively. | 9 | Molecular subtyping and genomic profiling expand precision medicine in refractory metastatic triple-negative breast cancer:the FUTURE trial显示文摘Triple-negative breast cancer(TNBC)is a highly heterogeneous disease,and molecular subtyping may result in improved diagnostic precision and targeted therapies.Our previous study classified TNBCs into four subtypes with putative therapeutic targets.Here,we conducted the FUTURE trial(ClinicalTrials.gov identifer:NCT03805399),a phaseⅠb/Ⅱsubtyping-based and genomic biomarker-guided umbrella trial,to evaluate the efficacy of these targets.Patients with refractory metastatic TNBC were enrolled and stratifed by TNBC subtypes and genomic biomarkers,and assigned to one of these seven arms:(A)pyrotinib with capecitabine,(B)androgen receptor inhibitor with CDK4/6 inhibitor,(C)anti PD-1 with nab-paclitaxel,(D)PARP inhibitor included,(E)and(F)anti-VEGFR included,or(G)mTOR inhibitor with nab paclitaxel.The primary end point was the objective response rate(ORR).We enrolled 69 refractory metastatic TNBC patients with a median of three previous lines of therapy(range,1-8).Objective response was achieved in 20(29.0%,95%confidence interval(CI):18.7%-41.2%)of the 69 intention-to-treat(ITT)patients.Our results showed that immunotherapy(arm C),in particular,achieved the highest ORR(52.6%,95%CI:28.9%-75.6%)in the ITT population.Arm E demonstrated favorable ORR(26.1%,95%C:10.2%-48.4%in the ITT population)but with more high grade(23)adverse events.Somatic mutations of TOP2A and CD8 immunohistochemical score may have the potential to predict immunotherapy response in the immunomodulatory subtype of TNBC.In conclusion,the phaseⅠb/ⅡFUTURE trial suggested a new concept for TNBC treatment,demonstrating the clinical beneft of subtyping-based targeted therapy for refractory metastatic TNBC. | Yi-Zhou Jiang Yin Liu Yi Xiao Xin Hu Lin Jiang Wen-ia Zuo Ding Ma Jiahan Ding Xiaoyu Zhu Jianjun Zou Claire Verschraegen Daniel G.Stover Virginia Kaklamani Zhong-Hua Wang Zhi-Ming Shao | 2021 | Cell Research2021,31,2: | 37 |
| 10 | Paleozoic and Mesozoic Basement Magmatisms of Eastern Qaidam Basin,Northern Qinghai-Tibet Plateau:LA-ICP-MS Zircon U-Pb Geochronology and its Geological Significance显示文摘The eastern margin of the Qaidam Basin lies in the key tectonic location connecting the Qinling,Qilian and East Kunlun orogens.The paper presents an investigation and analysis of the geologic structures of the area and LA-ICP MS zircon U-Pb dating of Paleozoic and Mesozoic magmatisms of granitoids in the basement of the eastern Qaidam Basin on the basis of 16 granitoid samples collected from the South Qilian Mountains,the Qaidam Basin basement and the East Kunlun Mountains.According to the results in this paper,the basement of the basin,from the northern margin of the Qaidam Basin to the East Kunlun Mountains,has experienced at least three periods of intrusive activities of granitoids since the Early Paleozoic,i.e.the magmatisms occurring in the Late Cambrian(493.1±4.9 Ma),the Silurian(422.9±8.0 Ma-420.4±4.6 Ma) and the Late Permian-Middle Triassic(257.8±4.0 Ma-228.8±1.5 Ma),respectively.Among them,the Late Permian-Middle Triassic granitoids form the main components of the basement of the basin.The statistics of dated zircons in this paper shows the intrusive magmatic activities in the basement of the basin have three peak ages of 244 Ma(main),418 Ma,and 493 Ma respectively.The dating results reveal that the Early Paleozoic magmatism of granitoids mainly occurred on the northern margin of the Qaidam Basin and the southern margin of the Qilian Mountains,with only weak indications in the East Kunlun Mountains.However,the distribution of Permo-Triassic(P-T) granitoids occupied across the whole basement of the eastern Qaidam Basin from the southern margin of the Qilian Mountains to the East Kunlun Mountains.An integrated analysis of the age distribution of P-T granitoids in the Qaidam Basin and its surrounding mountains shows that the earliest P-T magmatism(293.6-270 Ma) occurred in the northwestern part of the basin and expanded eastwards and southwards,resulting in the P-T intrusive magmatism that ran through the whole basin basement.As the Cenozoic basement thrust system developed in the eastern Qaidam Basin,the nearly N-S-trending shortening and deformation in the basement of the basin tended to intensify from west to east,which went contrary to the distribution trend of N-S-trending shortening and deformation in the Cenozoic cover of the basin,reflecting that there was a transformation of shortening and thickening of Cenozoic crust between the eastern and western parts of the Qaidam Basin,i.e.,the crustal shortening of eastern Qaidam was dominated by the basement deformation(triggered at the middle and lower crust),whereas that of western Qaidam was mainly by folding and thrusting of the sedimentary cover(the upper crust). | CHEN Xuanhua George GEHRELS YIN An LI Li JIANG Rongbao | 2012 | Acta Geologica Sinica(English Edition)2012,86,2: | 32 |
| 11 | Volumetric-modulated arc therapy vs c-IMRT in esophageal cancer:A treatment planning comparison显示文摘AIM:To compare the volumetric-modulated arc therapy AT plans ith conventional sliding indo intensity-modulated radiotherapy c-I RT plans in esophageal cancer EC . METHODS:Tenty patients ith EC ere selected, including 5 cases located in the cervical, the upper, the middle and the lo er thorax, respectively. Five plans ere generated ith the eclipse planning system:three using c-IMRT with 5 fields (5F), 7 fields (7F) and 9 fields (9F), and two using VMAT with a single arc 1A and double arcs 2A . The treatment plans ere designed to deliver a dose of 60 Gy to the plan-ning target volume Tith the same constrains in a 2.0 Gy daily fraction, 5 d a eek. lans ere normalized to 95% of the T that received 100% of the prescribed dose. We examined the dose-volume histogram parameters of T and the organs at risk OAR such as lungs, spinal cord and heart. onitor units U and normal tissue complication probability NTC of OAR ere also reported. RESULTS:Both c-I RT and AT plans resulted in abundant dose coverage of T for EC of different locations. The dose conformity to T as improved as the number of field in c-IMRT or rotating arc in VMAT as increased. The doses to T and OAR in AT plans ere not statistically different in comparison ith c-I RT plans, ith the follo ing exceptions:in cervical and upper thoracic EC, the conformity index CI as higher in VMAT (1A 0.78 and 2A 0.8) than in c-IMRT (5F 0.62, 7F 0.66 and 9F 0.73) and homogeneity was slightly better in c-IMRT (7F 1.09 and 9F 1.07) than in VMAT (1A 1.1 and 2A 1.09). Lung V30 was lower in VMAT (1A 12.52 and 2A 12.29) than in c-IMRT (7F 14.35 and 9F 14.81). The humeral head doses were significantly increased in AT as against c-I RT. | 15 | Trends in cancer mortality in China from 2004 to 2018:A nationwide longitudinal study显示文摘Background:The long-term trend in cancer death in a rapidly developing country provides information for cancer prophylaxis.Here,we aimed to identify the trends in cancer mortality in China during the 2004-2018 period.Methods:Using raw data from the national mortality surveillance system of China,we assessed the mortalities of all cancer and site-specific cancers during the 2004-2018 period.The participants were divided into three age groups:≥65 years,40-64 years,and≤39 years.Changing trends in cancer death by gender,residency,and tumor location were estimated using fitting joinpoint models to log-transformed crude mortality rates(CMRs)and age-standardizedmortality rates(ASMRs).Results:Cancer death accounted for 24% of all-cause of death in China during 2014-2018.The CMR of all cancer was 150.0 per 100,000 persons.Cancer was the leading cause of death in the population<65 years.The six major cancer types(lung/bronchus cancer,liver cancer,stomach cancer,esophagus cancer,colorectal cancer,and pancreas cancer)accounted for 75.85% of all cancer deaths.The CMR of all cancer increased while the ASMR decreased during 2014-2018(P<0.001).Lung/bronchus cancer and liver cancer were the leading causes of cancer death in the population<65 years,accounting for 45.31%(CMR)and 44.35%(ASMR)of all cancer death,respectively.The ASMR of liver cancer was higher in the 40-64 years population than in the≥65 years population,in contrast to the other five major cancers.The ASMRs of liver cancer,stomach cancer,and esophagus cancer decreased although they were higher in rural residents than in urban residents;the ASMRs of lung/bronchus cancer,colorectal cancer,and pancreas cancer increased in rural residents although they were higher in urban residents than in rural residents during 2014-2018.Conclusion:Although the ASMR of all cancer decreased in China during 2004-2018,lung/bronchus cancer and liver cancer remained the leading causes of cancer-related premature death.Lung/bronchus cancer,colorectal cancer,and pancreas cancer increased in rural residents. | Dongming Jiang Lijuan Zhang Wenbin Liu Yibo Ding Jianhua Yin Rongbing Ren Qi Li Yifan Chen Jiaying Shen Xiaojie Tan Hongwei Zhang Guangwen Cao | 2021 | Cancer Communications2021,41,10: | 23 |

When liver fibrosis is not well controlled, it can progress to liver cirrhosis, but it is reversible in principle. Thus far, no efficient therapy is available for treatment of liver fibrosis. Although liver transplantation is the preferred strategy, there are many challenges remaining in this approach, such as shortage of donor organs, immunological rejection, and surgical complications. Hence, there is a great need for an alternative therapeutic strategy. Currently, mesenchymal stem cell (MSC) therapy is considered a promising therapeutic strategy for the treatment of liver fibrosis;advantageously, the characteristics of MSCs are continuous self-renewal, proliferation, multipotent differentiation, and immunomodulatory activities. The human umbilical cord-derived (hUC)-MSCs possess not only the common attributes of MSCs but also more stable biological characteristics, relatively easy accessibility, abundant source, and no ethical issues (e.g., bone marrow being the adult source), making hUC-MSCs a good choice for treatment of liver fibrosis. In this review, we summarize the biological characteristics of hUC-MSCs and their paracrine effects, exerted by secretion of various cytokines, which ultimately promote liver repair through several signaling pathways. Additionally, we discuss the capacity of hUC-MSCs to differentiate into hepatocyte-like cells for compensating the function of existing hepatocytes, which may aid in amelioration of liver fibrosis.
