| 2 | Regulatory effect of gut microbes on blood pressure显示文摘Hypertension is an important global public health issue because of its high morbidity as well as the increased risk of other diseases.Recent studies have indicated that the development of hypertension is related to the dysbiosis of the gut microbiota in both animals and humans.In this review,we outline the interaction between gut microbiota and hypertension,including gut microbial changes in hypertension,the effect of microbial dysbiosis on blood pressure(BP),indicators of gut microbial dysbiosis in hypertension,and the microbial genera that affect BP at the taxonomic level.For example,increases in Lactobacillus,Roseburia,Coprococcus,Akkermansia,and Bifidobacterium are associated with reduced BP,while increases in Streptococcus,Blautia,and Prevotella are associated with elevated BP.Furthermore,we describe the potential mechanisms involved in the regulation between gut microbiota and hypertension.Finally,we summarize the commonly used treatments of hypertension that are based on gut microbes,including fecal microbiota transfer,probiotics and prebiotics,antibiotics,and dietary supplements.This review aims to find novel potential genera for improving hypertension and give a direction for future studies on gut microbiota in hypertension. | Dong Yan Ye Sun Xiaoyue Zhou Wenhao Si Jieyu Liu Min Li Minna Wu | 2022 | Animal Models and Experimental Medicine2022,5,6: | 1 |
| 3 | Science with the 2.5-meter Wide Field Survey Telescope(WFST)显示文摘The Wide Field Survey Telescope(WFST) is a dedicated photometric surveying facility being built jointly by University of Science and Technology of China(USTC) and the Purple Mountain Observatory(PMO). It is equipped with a 2.5-meter diameter primary mirror, an active optics system, and a mosaic CCD camera with 0.73 gigapixels on the primary focal plane for highquality image capture over a 6.5-square-degree field of view. The installation of WFST near the summit of Saishiteng mountain in the Lenghu region is scheduled in summer of 2023, and the operation is planned to start three months later. WFST will scan the northern sky in four optical bands(u, g, r and i) at cadences from hourly/daily in the deep high-cadence survey(DHS) program, to semi-weekly in the wide field survey(WFS) program. During a photometric night, a nominal 30 s exposure in the WFS program will reach a depth of 22.27, 23.32, 22.84, and 22.31(AB magnitudes) in these four bands, respectively, allowing for the detection of a tremendous amount of transients in the low-z universe and a systematic investigation of the variability of Galactic and extragalactic objects. In the DHS program, intranight 90 s exposures as deep as 23(u) and 24 mag(g), in combination with target of opportunity follow-ups, will provide a unique opportunity to explore energetic transients in demand for high sensitivities, including the electromagnetic counterparts of gravitational wave events, supernovae within a few hours of their explosions,tidal disruption events and fast, luminous optical transients even beyond redshift of unity. In addition, the final 6-year co-added images, anticipated to reach g■25.8 mag in WFS or 1.5 mags deeper in DHS, will be of fundamental importance to general Galactic and extragalactic science. The highly uniform legacy surveys of WFST will serve as an indispensable complement to those of the Vera C. | 9 | KDELR2 promotes breast cancer proliferation via HDAC3-mediated cell cycle progression显示文摘Background:Histone deacetylases(HDACs)engage in the regulation of various cellular processes by controlling global gene expression.The dysregulation of HDACs leads to carcinogenesis,making HDACs ideal targets for cancer therapy.However,the use of HDAC inhibitors(HDACi)as single agents has been shown to have limited success in treating solid tumors in clinical studies.This study aimed to identify a novel downstream effector of HDACs to provide a potential target for combination therapy.Methods:Transcriptome sequencing and bioinformatics analysis were performed to screen for genes responsive toHDACi in breast cancer cells.The effects of HDACi on cell viability were detected using the MTT assay.The mRNA and protein levels of genes were determined by quantitative reverse transcription-PCR(qRT-PCR)andWestern blotting.Cell cycle distribution and apoptosis were analyzed by flow cytometry.The binding of CREB1(cAMP-response element binding protein 1)to the promoter of the KDELR(The KDEL(Lys-Asp-Glu-Leu)receptor)gene was validated by the ChIP(chromatin immunoprecipitation assay).The association between KDELR2 and protein of centriole 5(POC5)was detected by immunoprecipitation.A breast cancer-bearing mouse model was employed to analyze the effect of the HDAC3-KDELR2 axis on tumor growth.Results:KDELR2 was identified as a novel target of HDAC3,and its aberrant expression indicated the poor prognosis of breast cancer patients.We found a strong correlation between the protein expression patterns of HADC3 and KDELR2 in tumor tissues from breast cancer patients.The results of the ChIP assay and qRT-PCR analysis validated that HDAC3 transactivated KDELR2 via CREB1.The HDAC3-KDELR2 axis accelerated the cell cycle progression of cancer cells by protecting the centrosomal protein POC5 from proteasomal degradation.Moreover,the HDAC3-KDELR2 axis promoted breast cancer cell proliferation and tumorigenesis in vitro and in vivo.Conclusion:Our results uncovered a previously unappreciated function of KDELR2 in tumorigenesis,linking a critical Golgi-the endoplasmic reticulum traffic transport protein to HDAC-controlled cell cycle progression on the path of cancer development and thus revealing a potential therapeutical target for breast cancer. | Haoran Wei Wenhao Ma Xiaofei Lu Haiying Liu Kashuai Lin Yinghui Wang Zijian Ye Linchong Sun Zhitong Huang Tingting Pan Zilong Zhou Eric Y.Cheng Huafeng Zhang Ping Gao Xiuying Zhong | 2021 | Cancer Communications2021,41,9: | 0 |

| 11 | Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND experiment. | 16 | Systematic Genome-Wide Profiles Reveal Alternative Splicing Landscape and Implications of Splicing Regulator DExD-Box Helicase 21 in Aggressive Progression of Adrenocortical Carcinoma显示文摘Alternative splicing(AS)in the tumor biological process has provided a novel perspective on carcinogenesis.However,the clinical significance of individual AS patterns of adrenocortical carcinoma(ACC)has been underestimated,and in-depth investigations are lacking.We selected 76 ACC samples from the Cancer Genome Atlas(TCGA)SpliceSeq and SpliceAid2 databases,and 39 ACC samples from Fudan University Shanghai Cancer Center(FUSCC).Prognosis-related AS events(PASEs)and survival analysis were evaluated based on prediction models constructed by machine-learning algorithm.In total,23,984 AS events and 3,614 PASEs were detected in the patients with ACC.The predicted risk score of each patient suggested that eight PASEs groups were significantly correlated with the clinical outcomes of these patients(p<0.001).Prognostic models produced AUC values of 0.907 in all PASEs’groups.Eight splicing factors(SFs),including BAG2,CXorf56,DExD-Box Helicase 21(DDX21),HSPB1,MBNL3,MSI1,RBMXL2,and SEC31B,were identified in regulatory networks of ACC.DDX21 was identified and validated as a novel clinical promoter and therapeutic target in 115 patients with ACC from TCGA and FUSCC cohorts.In conclusion,the strict standards used in this study ensured the systematic discovery of profiles of AS events using genome-wide cohorts.Our findings contribute to a comprehensive understanding of the landscape and underlying mechanism of AS,providing valuable insights into the potential usages of DDX21 for predict-ing prognosis for patients with ACC. | Wenhao Xu Aihetaimujiang Anwaier Wangrui Liu Xi Tian Wen-Kai Zhu Jian Wang Yuanyuan Qu Hailiang Zhang Dingwei Ye | 2021 | Phenomics2021,1,6: | 0 |
