2Focal adhesion protein Kindlin-2 regulates bone homeostasis in mice显示文摘Our recent studies demonstrate that the focal adhesion protein Kindlin-2 is critical for chondrogenesis and early skeletal development. Here, we show that deleting Kindlin-2 from osteoblasts using the 2.3-kb mouse Col1 a1-Cre transgene minimally impacts bone mass in mice, but deleting Kindlin-2 using the 10-kb mouse Dmp1-Cre transgene, which targets osteocytes and mature osteoblasts, results in striking osteopenia in mice. Kindlin-2 loss reduces the osteoblastic population but increases the osteoclastic and adipocytic populations in the bone microenvironment. Kindlin-2 loss upregulates sclerostin in osteocytes,downregulates β-catenin in osteoblasts, and inhibits osteoblast formation and differentiation in vitro and in vivo. Upregulation ofβ-catenin in the mutant cells reverses the osteopenia induced by Kindlin-2 deficiency. Kindlin-2 loss additionally increases the expression of RANKL in osteocytes and increases osteoclast formation and bone resorption. Kindlin-2 deletion in osteocytes promotes osteoclast formation in osteocyte/bone marrow monocyte cocultures, which is significantly blocked by an anti-RANKLneutralizing antibody. 5Fusaricidins in Paenibacillus polymyxa A21 and their antagonistic activity against Botrytis cinerea on tomato显示文摘Four kinds of antifungal compounds from an extract of Paenibacillus polymyxa A21 with molecular masses of 883.56, 897.59, 947.55, and 961.58 Da were characterized as the members of fusaricidin-type of antibiotics according to LC-MS analysis. Fusaricidins isolated from culture filtrate displayed high antagonistic activity against several plant fungal pathogens, especially Botrytis cinerea, the causal agent of gray mold. The fusaricidins biosynthetic gene cluster(BGC) from A21 was cloned by PCR and comparative cluster analysis revealed that gene fus TE, the 3′ boundary of the fusaricidin BGC in strain PKB1, was not present in fusaricidin BGC of A21, indicating that fus TE is not necessary for fusaricidin synthesis. Fusaricidin extract from A21 significantly reduced gray mold disease incidence and severity on tomato. The m RNA levels for three pathogenesis-related proteins(PRs) revealed that treatment of tomato leaves with fusaricidin extract induced the expression of PR genes to different levels, suggesting that one reason for the reduction of gray mold infection by fusaricidin is induction of PR proteins, which lead to increased resistance to pathogens. 6A novel sprayable thermosensitive hydrogel coupled with zinc modified metformin promotes the healing of skin wound显示文摘A novel sprayable adhesive is established(ZnMet-PF127)by the combination of a thermosensitive hydrogel(Pluronic F127,PF127)and a coordination complex of zinc and metformin(ZnMet).Here we demonstrate that ZnMet-PF127 potently promotes the healing of traumatic skin defect and burn skin injury by promoting cell proliferation,angiogenesis,collagen formation.Furthermore,we find that ZnMet could inhibit reactive oxygen species(ROS)production through activation of autophagy,thereby protecting cell from oxidative stress induced damage and promoting healing of skin wound.ZnMet complex exerts better effects on promoting skin wound healing than ZnCl2 or metformin alone.ZnMet complex also displays excellent antibacterial activity against Staphylococcus aureus or Escherichia coli,which could reduce the incidence of skin wound infections.Collectively,we demonstrate that sprayable PF127 could be used as a new drug delivery system for treatment of skin injury.The advantages of this sprayable system are obvious:(1)It is convenient to use;(2)The hydrogel can cover irregular skin defect sites evenly in a liquid state.In combination with this system,we establish a novel sprayable adhesive(ZnMet-PF127)and demonstrate that it is a potential clinical treatment for traumatic skin defect and burn skin injury.Zhengwei Liu Wanze Tang Jiayi Liu Yingying Han Qinnan Yan Yuechao Dong Xiaomei Liu Dazhi Yang Guixing Ma Huiling Cao 2023Bioactive Materials2023,,2:3
7Reactive MnO_(2) template-assisted synthesis of double-shelled PPy hollow nanotubes to boost microwave absorption显示文摘Microwave absorbing materials(MAMs)with wide effective absorption bandwidth(EAB)and low filling ratio are highly desirable for practical applications.Rational design in components and structures is one of the effective strategies to achieve MAMs with high performance.Herein,double-shelled hollow(DSH)polypyrrole(PPy)nanotubes were synthesized with hydrochloric acid(HCl)and sodium pstyrene sulfonate(SS)co-doping polymerization process using manganese dioxide(MnO_(2))nanorods as a self-sacrifice template.With the increase of HCl concentration,the 1D MnO_(2) core diminishes gradually to form the MnO_(2)@PPy coaxial nanostructures and finally the DSH PPy nanotube,which tunes the microwave absorption performance.Importantly,the DSH PPy nanotubes exhibit excellent microwave absorption of an optimal reflection loss of–50.4 dB and a wide EAB of 7.7 GHz with a low filling ratio of 5 wt%in a paraffin wax matrix.The excellent microwave absorption is believed to be mainly attributed to the enhanced synergistic effects of interfacial polarization and conduction loss arising from the unique DSH structure and the co-doping polymerization.Huiling Gu Ji Huang Na Li Hua Yang Gang Chen Chengjun Dong Chunhong Gong Hongtao Guan 2023Journal of Materials Science & Technology2023,,15:3

10Co-occurrence of genes for antibiotic resistance and arsenic biotransformation in paddy soils显示文摘Paddy soils are potential hotspots of combined contamination with arsenic(As) and antibiotics, which may induce co-selection of antibiotic resistance genes(ARGs) and As biotransformation genes (ABGs), resulting in dissemination of antimicrobial resistance and modification in As biogeochemical cycling. So far, little information is available for these coselection processes and specific patterns between ABGs and ARGs in paddy soils. Here, the16S rRNA amplicon sequencing and high-throughput quantitative PCR and network analysis were employed to investigate the dynamic response of ABGs and ARGs to As stress and manure application. The results showed that As stress increased the abundance of ARGs and mobile genetic elements (MGEs), resulting in dissemination risk of antimicrobial resistance. Manure amendment increased the abundance of ABGs, enhanced As mobilization and methylation in paddy soil, posing risk to food safety. The frequency of the co-occurrence between ABGs and ARGs, the host bacteria carrying both ARGs and ABGs were increased by As or manure treatment, and remarkably boosted in soils amended with both As and manure. Multidrug resistance genes were found to have the preference to be co-selected with ABGs, which was one of the dominant co-occurring ARGs in all treatments, and manure amendment increased the frequency of Macrolide-Lincosamide-Streptogramin B resistance(MLSB) to co-occur with ABGs. Bacillus and Clostridium of Firmicutes are the dominant host bacteria carrying both ABGs and ARGs in paddy soils. 16Extraction,separation and refining of microcrystalline cellulose from wheat straw using various pretreatments显示文摘This study focused on the manufacture of microcrystalline cellulose(MCC)from wheat straw using environmentally-friendly solvents.Raw cellulose was separated from wheat straw after thermal decomposition of lignin followed by dissolution of lignin using a recyclable ethanol/acetic acid/water solvent system.Then,pure cellulose was produced using a four-step refining process,including chelating,O_(3),H_(2)O_(2),and xylanase treatments.Finally,MCC was obtained through hydrolysis,drying,and mechanical treatments.The effects of acetic acid,O_(3),H_(2)O_(2),NaOH,pretreatment time,and temperature on the properties of wheat straw cellulose(including Kappa number,yield,α-cellulose content,crystallinity,KMnO4 value,degree of polymerization(DP),and brightness)were investigated.The results showed that the addition of acetic acid enhanced lignin removal and hemicellulose degradation,improving the purity of the raw cellulose.The optimized acetic acid dosage in the wheat straw thermal decomposition step was 2%(w/w).The optimized O_(3)dosage was 1.2%(w/w).The optimized conditions for H_(2)O_(2)treatment were found to be 3%(w/w)H_(2)O_(2)and 1.8%(w/w)NaOH at 70°C for 120 min.The KMnO4 value was 2.0,brightness was 84.1%ISO,the viscosity was 934 mL/g,and the DP was 626 for refined cellulose.Xylanase effectively improved theα-cellulose content of wheat straw cellulose.With an optimized xylanase dosage of 1.5 IU/g,theα-cellulose content was 94.7%,the brightness was 85.6%ISO,and the DP was 615 for wheat straw cellulose.Li Jinbao Dong Huiling Xiu Huijuan Zhang Meiyun Kamireddy Srinivas Reddy Zhang Xuefei Ji Yun 2016International Journal of Agricultural and Biological Engineering2016,9,2:0
17Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND 18Current Effect of“4+7”Drug Centralized Volume Procurement and Suggestions for Future Improvement显示文摘Objective To explore the impact of the“4+7”city drug centralized procurement policy implemented from April 1st,2019 on China’s pharmaceutical market and the differences between this pilot scheme and the first volume procurement,so as to put forward some suggestions for this policy and improve the drug price formation mechanism.Methods The characteristics of the“4+7”volume procurement policy and its improvements were summarized through literature research,and the distribution of the winning enterprises in volume procurement was shown by the chart.Then the overall situation of pharmaceutical enterprises in China was analyzed.Results and Conclusion According to the main distribution of enterprises in the alliance region,we can see that the first round of bid-winning enterprises with 25 varieties are mainly distributed in the eastern coastal cities of Jiangsu,Zhejiang,Shandong,and Guangdong,which shows a trend of developing to provinces of Sichuan,Hubei,and Hunan.With the increase of volume procurement inland cities will be included in the future.This pilot scheme of volume procurement is a reform of the entire pharmaceutical industry.It has improved the market structure and promoted the transformation and upgrading of regional medicine representatives.The most important thing is to enable the public to better enjoy the benefits brought by the low-priced drugs.However,for some pharmaceutical enterprises and drug retail industry,it is a challenge.Therefore,it is necessary to improve the policy and strengthen market supervision and management.Piao Huiling Wei Xingchen Wang Nan Wang Xing Zhou Yusheng Dong Li 2020Asian Journal of Social Pharmacy2020,15,4:0
19The role of E3 ubiquitin ligases in bone homeostasis and related diseases显示文摘The ubiquitin-proteasome system(UPS)dedicates to degrade intracellular proteins to modulate demic homeostasis and functions of organisms.These enzymatic cascades mark and modifies target proteins diversly through covalently binding ubiquitin molecules.In the UPS,E3 ubiquitin ligases are the crucial constituents by the advantage of recognizing and presenting proteins to proteasomes for proteolysis.As the major regulators of protein homeostasis,E3 ligases are indispensable to proper cell manners in diverse systems,and they are well described in physiological bone growth and bone metabolism.Pathologically,classic bone-related diseases such as metabolic bone diseases,arthritis,bone neoplasms and bone metastasis of the tumor,etc.,were also depicted in a UPS-dependent manner.Therefore,skeletal system is versatilely regulated by UPS and it is worthy to summarize the underlying mechanism.Furthermore,based on the current status of treatment,normal or pathological osteogenesis and tumorigenesis elaborated in this review highlight the clinical significance of UPS research.As a strategy possibly remedies the limitations of UPS treatment,emerging PROTAC was described comprehensively to illustrate its potential in clinical application.Altogether,the purpose of this review aims to provide more evidence for exploiting novel therapeutic strategies based on UPS for bone associated diseases.Yuechao Dong Yangshan Chen Guixing Ma Huiling Cao 2023Acta Pharmaceutica Sinica B2023,13,10:0
20Microglial Depletion does not Afect the Laterality of Mechanical Allodynia in Mice显示文摘Mechanical allodynia(MA),including punctate and dynamic forms,is a common and debilitating symptom suffered by millions of chronic pain patients.Some peripheral injuries result in the development of bilateral MA,while most injuries usually led to unilateral MA.To date,the control of such laterality remains poorly understood.Here,to study the role of microglia in the control of MA laterality,we used genetic strategies to deplete microglia and tested both dynamic and punctate forms of MA in mice.Surprisingly,the depletion of central microglia did not prevent the induction of bilateral dynamic and punctate MA.Moreover,in dorsal root ganglion-dorsal root-sagittal spinal cord slice preparations we recorded the low-threshold Aβ-fiber stimulation-evoked inputs and outputs of superficial dorsal horn neurons.Consistent with behavioral results,microglial depletion did not prevent the opening of bilateral gates for Aβpathways in the superficial dorsal horn.This study challenges the role of microglia in the control of MA laterality in mice.Future studies are needed to further understand whether the role of microglia in the control of MA laterality is etiology-or species-specific.Quan Ma Dongmei Su Jiantao Huo Guangjuan Yin Dong Dong Kaifang Duan Hong Cheng Huiling Xu Jiao Ma Dong Liu Bin Mou Jiyun Peng Longzhen Cheng 2023Neuroscience Bulletin2023,39,8:0
