A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. Further efforts should focus on identifying the etiology of the SARS-associated virus and ruling out conclusively the existence of otherpossible SARS-related pathogen(s). | QIN E'de ZHU Qingyu YU Man FAN Baochang CHANG Guohui SI Bingyin YANG Bao PENG Wenming JIANG Tao LIU Bohua DENG Yongqiang LIU Hong ZHANG Yu WANG Cui LI Yuquan GAN Yonghua LI Xiaoyu L Fushuang TAN Gang CAO Wuchun, YANG Ruifu Institute of Microbiology and Epidemiology, Chinese Academy of Military Medical Sciences, Beijing 100071, China WANG Jian, LI Wei, XU Zuyuan, LI Yan, WU Qingfa, LIN Wei, CHEN Weijun, TANG Lin, DENG Yajun, HAN Yujun, LI Changfeng, LEI Meng, LI Guoqing, LI Wenjie, L Hong, SHI Jianping, TONG Zongzhong, ZHANG Feng, LI Songgang, LIU Bin, LIU Siqi, DONG Wei, WANG Jun, Gane K-S Wong, YU Jun & YANG Huanming* Beijing Genomics Institute, Chinese Academy of Sciences, Beijing 101300 National Center for Genome Information, Beijing 101300, China | 2003 | Chinese Science Bulletin2003,48,10: | 121
Drug nanoclusters formed in confined nano-cages of CD-MOF: dramatic enhancement of solubility and bioavailability of azilsartan显示文摘Tremendous efforts have been devoted to the enhancement of drug solubility using nanotechnologies, but few of them are capable to produce drug particles with sizes less than a few nanometers. This challenge has been addressed here by using biocompatible versatile γ-cyclodextrin(γ-CD) metal-organic framework(CD-MOF) large molecular cages in which azilsartan(AZL) was successfully confined producing clusters in the nanometer range. This strategy allowed to improve the bioavailability of AZL in Sprague–Dawley rats by 9.7-fold after loading into CD-MOF. The apparent solubility of AZL/CD-MOF was enhanced by 340-fold when compared to the pure drug. Based on molecular modeling, a dual molecular mechanism of nanoclusterization and complexation of AZL inside the CD-MOF cages was proposed, which was confirmed by small angle X-ray scattering(SAXS) and synchrotron radiation-Fourier transform infrared spectroscopy(SR-FTIR) techniques. In a typical cage-like unit of CD-MOF, three molecules of AZL were included by the γ-CD pairs, whilst other three AZL molecules formed a nanocluster inside the 1.7 nm sized cavity surrounded by six γ-CDs. This research demonstrates a dual molecular mechanism of complexation and nanoclusterization in CD-MOF leading to significant improvement in the bioavailability of insoluble drugs. | Yuanzhi He Wei Zhang Tao Guo Guoqing Zhang Wei Qin Liu Zhang Caifen Wang Weifeng Zhu Ming Yang Xiaoxiao Hu Vikramjeet Singh Li Wu Ruxandra Gref Jiwen Zhang | 2019 | Acta Pharmaceutica Sinica B2019,9,1: | 13
Geological Controls on the CBM Productivity of No.15 Coal Seam of Carboniferous–Permian Taiyuan Formation in Southern Qinshui Basin and Prediction for CBM High-yield Potential Regions显示文摘Coalbed methane(CBM) resources in No.15 coal seam of Taiyuan Formation account for 55% of the total CBM resources in southern Qinshui Basin(SQB), and have a great production potential. This study aims at investigating the CBM production in No.15 coal seam and its influence factors. Based on a series of laboratory experiments and latest exploration and development data from local coal mines and CBM companies, the spatial characteristics of gas production of No.15 coal seam were analyzed and then the influences of seven factors on the gas productivity of this coal seam were discussed, including coal thickness, burial depth, gas content, ratio of critical desorption pressure to original coal reservoir pressure(RCPOP), porosity, permeability, and hydrogeological condition. The influences of hydrological condition on CBM production were analyzed based on the discussions of four aspects: hydrogeochemistry, roof lithology and its distribution, hydrodynamic field of groundwater, and recharge rate of groundwater. Finally, a three-level analytic hierarchy process(AHP) evaluation model was proposed for predicting the CBM potentials of the No.15 coal seam in the SQB. The best prospective target area for CBM production of the No.15 coal seam is predicted to be in the districts of Panzhuang, Chengzhuang and south of Hudi. | ZHANG Zheng QIN Yong ZHUANG Xinguo LI Guoqing LIU Donghai | 2018 | Acta Geologica Sinica(English Edition)2018,92,6: | 5

Although one patient had platelet transfusion owing to Grade 4 thrombocytopenia in EL group, no significant difference (P = 0.637) were shown. The incidence of nausea/vomiting was higher in the EP group than in the EL group (96.7% vs 51.6%, P = 0.00). Conclusion: The EL regimen is an effective and low-toxicity chemotherapy and no inferior to EP regimen in treatment response, therefore, EL regimen maybe is a good choice for patients with extensive-stage SCLC. | Wanfeng Guo Guoqing Liao Hongjun Gao Hongmei Wang Chuanhao Tang Haifeng Qin Jianjie Li Xiaoyan Li Xiaoqing Liu | 2013 | The Chinese-German Journal of Clinical Oncology2013,12,8: | 2 |
Rapid and high precision measurement of opto-thermal relaxation with pump-probe method显示文摘Opto-thermal relaxation is one of the most important properties of nonlinear optical materials.Rapid and high precision measurement of this parameter is vital in both fundamental research and applications.Current measurement uses either complicated structure with poor precision or high power heating source with low efficiency. Here,we propose a pump-probe method(PPM) to optically measure the thermal relaxation using whispering gallery mode(WGM) microcavities. When the pump laser shines on a microcavity, the materials absorb the input power resonantly and heat up.Then the heat dissipates from the cavities to the surroundings. The opto-thermal effect induces a refractive index change reflected in the signal light transmission spectra.By analyzing the curve character of the transmission spectra of the signal response in the spontaneous relaxation process, the thermal relaxation time can be rapidly measured with high precision.Additionally,we systematically verify the PPM using microtoroids under various pump powers and at various locking points of the signal laser mode. The small rate of refractive index changes($10à8) can be discerned with an input pump power as low as 11.816 l W.Hence, the PPM can be used to detect refractive index perturbation, like gas or liquid sensing, temperature fluctuations with ultra-high sensitivity and be applied to optical materials analysis efficiently. | Tao Wang Xiao-Fei Liu Yunqi Hu Guoqing Qin Dong Ruan Gui-Lu Long | 2018 | Science Bulletin2018,63,5: | 2

Electron-ion collider in China显示文摘Lepton scattering is an established ideal tool for studying inner structure of small particles such as nucleons as well as nuclei. As a future high energy nuclear physics project,an Electron-ion collider in China(EicC)has been proposed. It will be constructed based on an upgraded heavy-ion accelerator,High Intensity heavy-ion Accelerator Facility(HIAF)which is currently under construction, together with a new electron ring. The proposed collider will provide highly polarized electrons(with a po-larization of 80%)and protons(with a polarization of 70%)with variable center of mass energies from 15 to 20 GeV and the luminosity of(2–3)×1033 cm^(−2)·s^(−1).Polarized deuterons and Helium-3,as well as unpolarized ion beams from Carbon to Uranium, will be also available at the EicC.The main foci of the EicC will be precision measurements of the structure of the nucleon in the sea quark region, including 3D tomography of nucleon;the partonic structure of nuclei and the parton interaction with the nuclear environment;the exotic states, especially those with heavy flavor quark contents. In addition, issues fundamental to understanding the origin of mass could be addressed by measurements of heavy quarkonia near-threshold production at the EicC.In order to achieve the above-mentioned physics goals,a hermetical detector system will be constructed with cutting-edge technologies. This document is the result of collective contributions and valuable inputs from experts across the globe. The EicC physics program complements the ongoing scientific programs at the Jefferson Laboratory and the future EIC project in the United States. The success of this project will also advance both nuclear and particle physics as well as accelerator and detector technology in China. | Daniele PAnderle Valerio Bertone Xu Cao Lei Chang Ningbo Chang Gu Chen Xurong Chen Zhuojun Chen Zhufang Cui Lingyun Dai Weitian Deng Minghui Ding Xu Feng Chang Gong Longcheng Gui Feng-Kun Guo Chengdong Han Jun He Tie-Jiun Hou Hongxia Huang Yin Huang KrešImir KumeričKi LPKaptari Demin Li Hengne Li Minxiang Li Xueqian Li Yutie Liang Zuotang Liang Chen Liu Chuan Liu Guoming Liu Jie Liu Liuming Liu Xiang Liu Tianbo Liu Xiaofeng Luo Zhun Lyu Boqiang Ma Fu Ma Jianping Ma Yugang Ma Lijun Mao Cédric Mezrag HervéMoutarde Jialun Ping Sixue Qin Hang Ren Craig DRoberts Juan Rojo Guodong Shen Chao Shi Qintao Song Hao Sun PawełSznajder Enke Wang Fan Wang Qian Wang Rong Wang Ruiru Wang Taofeng Wang Wei Wang Xiaoyu Wang Xiaoyun Wang Jiajun Wu Xinggang Wu Lei Xia Bowen Xiao Guoqing Xiao Ju-Jun Xie Yaping Xie Hongxi Xing Hushan Xu Nu Xu Shusheng Xu Mengshi Yan Wenbiao Yan Wencheng Yan Xinhu Yan Jiancheng Yang Yi-Bo Yang Zhi Yang Deliang Yao Zhihong Ye Peilin Yin C-PYuan Wenlong Zhan Jianhui Zhang Jinlong Zhang Pengming Zhang Yifei Zhang Chao-Hsi Chang Zhenyu Zhang Hongwei Zhao Kuang-Ta Chao Qiang Zhao Yuxiang Zhao Zhengguo Zhao Liang Zheng Jian Zhou Xiang Zhou Xiaorong Zhou Bingsong Zou Liping Zou | 2021 | Frontiers of physics2021,16,6: | 1

Bio-clickable mussel-inspired peptides improve titanium-based material osseointegration synergistically with immunopolarization-regulation显示文摘Upon the osteoporotic condition, sluggish osteogenesis, excessive bone resorption, and chronic inflammation make the osseointegration of bioinert titanium(Ti)implants with surrounding bone tissues difficult, often lead to prosthesis loosening, bone collapse, and implant failure. In this study,we firstly designed clickable mussel-inspired peptides(DOPA-N3)and grafted them onto the surfaces of Ti materials through robust catechol-TiO2 coordinative interactions. Then, two dibenzylcyclooctyne(DBCO)-capped bioactive peptides RGD and BMP-2 bioactive domain(BMP-2)were clicked onto the DOPA-N3-coated Ti material surfaces via bio-orthogonal reaction. We characterized the surface morphology and biocompatibility of the Ti substrates and optimized the osteogenic capacity of Ti surfaces through adjusting the ideal ratios of BMP-2/RGD at 3:1.In vitro, the dual-functionalized Ti substrates exhibited excellent promotion on adhesion and osteogenesis of mesenchymal stem cells(MSCs),and conspicuous immunopolarization-regulation to shift macrophages to alternative(M2)phenotypes and inhibit inflammation,as well as enhancement of osseointegration and mechanical stability in osteoporotic rats.In summary, our biomimetic surface modification strategy by bio-orthogonal reaction provided a convenient and feasible method to resolve the bioinertia and clinical complications of Ti-based implants, which was conducive to the long-term success of Ti implants, especially in the osteoporotic or inflammatory conditions. | Jie Sun Yingkang Huang Huan Zhao Junjie Niu Xuwei Ling Can Zhu Lin Wang Huilin Yang Zhilu Yang Guoqing Pan Qin Shi | 2022 | Bioactive Materials2022,7,3: | 0
Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector. Some of JUNO's features make it an excellent location for^8B solar neutrino measurements, such as its low-energy threshold, high energy resolution compared with water Cherenkov detectors, and much larger target mass compared with previous liquid scintillator detectors. In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable, assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition, approximately 60,000 signal and 30,000 background events are expected. This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter, which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle. Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better, depending on the central value, and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND experiment. | 14 | Genome-wide signatures of mammalian skin covering evolution显示文摘Animal body coverings provide protection and allow for adaptation to environmental pressures such as heat, ultraviolet radiation, water loss, and mechanical forces. Here, using a comparative genomics analysis of 39 mammal species spanning three skin covering types(hairless, scaly and spiny),we found some genes(e.g.,UVRAG,POLH,and XPC)involved in skin inflammation, skin innate immunity, and ultraviolet radiation damage repair were under selection in hairless ocean mammals(e.g.,whales and manatees). These signatures might be associated with a high risk of skin diseases from pathogens and ultraviolet radiation. Moreover, the genomes from three spiny mammal species shared convergent genomic regions(EPHB2,EPHA4,and NIN)and unique positively selected genes(FZD6,INVS,and CDC42)involved in skin cell polarity, which might be related to the development of spines. In scaly mammals, the shared convergent genomic regions(e.g.,FREM2)were associated with the integrity of the skin epithelium and epidermal adhesion. This study identifies potential convergent genomic features among distantly related mammals with the same skin covering type. | Peng Cao Qinlong Dai Cao Deng Xiang Zhao Shishan Qin Jian Yang Ran Ju Zhiwen Wang Guoqing Lu Xiaodong Gu Zhisong Yang Lifeng Zhu | 2021 | Science China(Life Sciences)2021,64,10: | 0
A unified representation method for interdisciplinary spatial earth data显示文摘Unified representation of spatial earth data is an essential scientific issue. The analysis and mining of interdisciplinary spatial earth data resources can help discover hidden scientific knowledge, and even reveal the intrinsic relationship among different disciplines. However, the different description methods and inner structures among interdisciplinary spatial earth data bring significant chal-lenges to unified data management and collaborative analysis in earth environment research. To address this issue, this paper pro-poses a unified representation method for interdisciplinary spatial earth data.First, this paper establishes a general metadata model and realizes the unified description of interdisciplinary data.Second,an entity data organization model is presented, which can realize the unified organization of entity data with different inner structures. Finally,we introduce the Spatial Earth Data Format(SEDF),a data format based on HDF5 for implementing the data organization model of interdisciplinary spatial earth data.Data representation experiments and validation are conducted to verify the availability and practicability of the proposed data representa-tion method. The results suggest the powerful ability to represent spatial earth data efficiently and ensure data integrity, which is convenient for data management and application. | Shuang Wang Jian Wang Qin Zhan Lianchong Zhang Xiaochuang Yao Guoqing Li | 2023 | Big Earth Data2023,7,1: | 0

In vitro degradation experiments indicated that POEDU nanospheres degraded faster than POEU nanospheres in mildly acidic and reductive environments. Doxorubicin(DOX) as a model antitumor drug was successfully incorporated into these nanospheres to give DOX-loaded nanoparticles(POEDU-DOX and POEU-DOX). In vitro drug release studies showed that release of DOX from dual-stimuli-sensitive POEDU-DOX was accelerated compared with release from the p H-sensitive POEU-DOX under DL-dithiothreitol(DTT) and mildly acidic conditions. In addition, in vitro uptake and cytotoxicity assays revealed that POEDU-DOX exhibited more efficient antitumor effect than POEU-DOX did against both twodimensional(2 D) cells and three-dimensional(3 D) multicellular tumor spheroids(MCTS). Finally, in a mice H22 tumor model,POEDU-DOX exhibited preferable antitumor capability. In conclusion, the pH and redox dual-stimuli-sensitive POEDU nanospheres can be superior drug carriers for cancer treatment. | Yan Huang Jiejie Qin Jun Wang Guoqing Yan Xin Wang Rupei Tang | 2018 | Science China Chemistry2018,61,11: | 0 |
Translation:Management of Coronavirus Disease 2019(COVID-19):Experience in Zhejiang Province,China显示文摘The current epidemic situation of coronavirus disease 2019(COVID-19)still remains severe. As the National Clinical Research Center for Infectious Diseases,The First Affiliated Hospital of the Zhejiang University School of Medicine is the primary medical care center for COVID-19 in Zhejiang Province.Based on the present expert consensus carried out by the National Health Commission and National Administration of Traditional Chinese Medicine, our team summarized and established an effective treatment strategy centered on“Four-Anti and Two-Balance”for clinical practice.The“Four-Anti and Two-Balance”strategy includes antivirus,anti-shock,antihypoxemia,and anti-secondary infection, and maintaining of water, electrolyte and acid/base balance and microecological balance.Simultaneously,an integrated multidisciplinary personalized treatment is recommended to improve therapeutic effects. The importance of early viral detection, dynamic monitoring of inflammatory indexes, and chest radiographs has been emphasized in clinical decision-making.Sputum was observed with the highest positive rate by reverse transcription-polymerase chain reaction(RTPRC).Viral nucleic acids could be detected in 10%of the patients’blood samples at the acute phase and 50%of patients had positive RT-PCR results in their feces. We also isolated live viral strains from feces, indicating potential infectiousness of feces.Dynamic cytokine detection was necessary to timely identify cytokine storms and for the application of the artificial liver blood purification system.The“Four-Anti and Two-Balance”strategy effectively increased cure rates and reduced mortality.Early antiviral treatment alleviated disease severity and prevented illness progression. We found that lopinavir/ritonavir combined with abidol showed antiviral effects against COVID-19.Shock and hypoxemia were usually caused by cytokine storms. The artificial liver blood purification system was able to rapidly remove inflammatory mediators and block the cytokine storm. Moreover,it also contributed to the balance of fluids, electrolytes, and acids/bases and thus improved treatment efficacy during critical illness. For cases of severe illness, early and also short periods of moderate glucocorticoid administration was supported.Patients with an oxygenation index below 200mmHg were transferred to the intensive care unit.Conservative oxygen therapy was preferred and noninvasive ventilation(NIV)was not recommended.Patients with mechanical ventilation were strictly supervised with cluster ventilator-associated pneumonia prevention strategies.Antimicrobial prophylaxis was prescribed rationally and was not recommended, except for patients with a long course of disease, repeated fever, and elevated procalcitonin, similarly secondary fungal infections were of concern. Some patients with COVID-19 showed intestinal microbial dysbiosis with decreased genus such as Lactobacillus and Bifidobacterium.Nutritional and gastrointestinal function should;therefore,be assessed for all patients.Nutritional support and application of prebiotics or probiotics were suggested to regulate the balance of intestinal microbiota and reduce the risk of secondary infections due to bacterial translocation.Anxiety and fear were common in patients with COVID-19.Therefore,we established a dynamic assessment and warning for psychological crises. We also integrated Chinese medicine in the treatment to promote rehabilitation. We optimized nursing processes for severe patients to promote their rehabilitation.Since viral clearance patterns after severe acute respiratory syndrome coronavirus 2 infections remained unclear,2 weeks quarantine for discharged patients was required, and a regular following-up was also needed. These Zhejiang experiences and suggestions have been implemented in our center and achieved good results. However, since COVID-19 was a newly emerging disease, more work is warranted to further improve strategies of prevention, diagnosis, and treatment for COVID-19. | Kaijin Xu Hongliu Cai Yihong Shen Qin Ni Yu Chen Shaohua Hu Jianping Li Huafen Wang Liang Yu He Huang Yunqing Qiu Guoqing Wei Qiang Fang Jianying Zhou Jifang Sheng Tingbo Liang Lanjuan Li | 2020 | Infectious Microbes & Diseases2020,2,2: | 0
Trends of SARS-CoV-2 Infection Among Couriers in Risky Business After the Optimization of Prevention and Control Measures——China,December 2022 to January 2023显示文摘Introduction:On December 7,2022,China implemented“Ten New Measures”to optimize prevention and control measures for coronavirus disease 2019(COVID-19).The purpose of this study was to evaluate the national and regional trends of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)infection among couriers in China from December 2022 to January 2023.Methods:Data from the National Sentinel Community-based Surveillance in China was utilized, including participants from 31 provincial-level administrative divisions and Xinjiang Production and Construction Corps.Participants were tested for SARS-CoV-2 infection twice a week from December 16,2022 to January 12,2023.Infection was defined as a positive result for SARS-CoV-2 nucleic acid or antigen. The daily average newly positive rate of SARSCoV-2 infection and the estimated daily percentage change(EDPC)were calculated. Results:In this cohort,8 rounds of data were collected. The daily average newly positive rate of SARS-CoV-2 infection decreased from 4.99%in Round 1 to 0.41%in Round 8,with an EDPC of−33.0%.Similar trends of the positive rate were also observed in the eastern(EDPC:−27.7%),central(EDPC:−38.0%)and western regions(EDPC:−25.5%).Couriers and community population showed a similar temporal trend, with the peak daily average newly positive rate of couriers being higher than that of community population. After Round 2,the daily average newly positive rate of couriers decreased sharply, becoming lower than that of community population in the same period. Conclusions:The peak of SARS-CoV-2 infection among couriers in China has passed. As couriers are a key population for SARS-CoV-2 infection, they should be monitored continuously. | Yu Wu Fan Ding Jue Liu Wenzhan Jing Wenxin Yan Chenyuan Qin Fangyuan Li Yanping Zhang Guoqing Shi Wannian Liang Qun Li Min Liu | 2023 | China CDC weekly2023,5,11: | 0
